摘 要:目的探讨脂多糖(LPS)对正常人肝内胆管上皮细胞(Hi BECs)中TLR4-NF-кB通路的影响。方法 Hi BECs常规细胞培养后,加入不同浓度的LPS(0.1、1、4、8、10μg/m L),刺激不同时间(3、6、9、12、24 h),用Realtime RT-PCR分析细胞内TLR4和NF-κB mRNA表达水平。结果 LPS刺激Hi BECs后TLR4和NF-κB mRNA表达水平均增加,Hi BECs在受到LPS(4μg/m L)刺激后TLR4和NF-κB mRNA表达水平随时间延长而增加,于6小时达高峰,之后下降。以相同时间(6 h)刺激Hi BECs后,TLR4和NF-κB mRNA表达水平随LPS浓度增高而增加,LPS浓度为4μg/m L时达高峰,之后下降。结论 LPS可以激活Hi BECs中的TLR4-NF-кB通路,并且有时效和量效依赖关系。Objective To investigate the effect of lipopolysaccharides ( LPS) on TLR4-NF-кB pathway in human intrahepatic biliary epithelial cells ( HiBECs) in vitro. Methods HiBECs were cultured and divided into normal control group ( NC) and LPS group. LPS group was treated with LPS at different time (3 h, 6 h, 9 h, 12 h, 24 h)and different concentrations (0. 1, 1, 4, 8, 10μg/mL). The expression of TLR4 and NF-кB was assessed by realtime-PCR and Western blot. Results LPS upregulated the expression of TLR4 and NF-кB in Hi-BECs. The TLR4 and NF-κB mRNA transcription was increased at different time of LPS stimulation, and reached its peak on 6 h and then gradually decreased. Simultaneously, the TLR4 and NF-κB mRNA expression level was increased in a dose-dependent manner,and reached its peak when used at 4 μg/mL LPS and then decreased. Conclusion LPS activates TLR4-NF-кB signaling in HiBECs in a time-and dose-dependent manner.
