摘 要:为了考察紫外线消毒对水中细菌的灭活效果及耐药性的影响,使用3种具有不同耐药性的大肠杆菌(大肠杆菌ATCC25922、对四环素和磺胺甲恶唑二重耐药的大肠杆菌SER2以及对四环素、磺胺甲恶唑、氨苄西林、庆大霉素、链霉素和头孢噻肟六重耐药的大肠杆菌SER6)进行紫外线消毒实验。通过平板计数法研究了不同剂量的紫外线辐照对大肠杆菌的灭活率,并利用K-B纸片琼脂扩散法和扫描电子显微镜分别考察了紫外线消毒后大肠杆菌耐药性和细胞形态的变化。结果表明,在20 m J/cm2的紫外线辐照剂量下大肠杆菌ATCC25922、SER2和SER6的对数灭活率分别为4.75、5.04和3.83。大肠杆菌ATCC25922和SER2的紫外线消毒灭活效果基本相同,而大肠杆菌SER6的灭活率偏低。经80 m J/cm2的紫外线消毒之后,大肠杆菌SER6对庆大霉素药敏纸片抑菌环直径较消毒前发生了显著变化(p<0.05)。高辐照剂量(200m J/cm2)的紫外线消毒能够造成菌株形态上的破坏,这是导致其灭活的原因之一。In order to investigate the inactivation of bacteria and their antibiotic-resistance variation by ultraviolet disinfection,three kinds of Escherichia coli( E. coli ATCC25922,tetracycline and sulfamethoxazole-resistant E. coli SER2 and tetracycline,sulfamethoxazole,ampicillin,gentamicin,streptomycin and cefotaximeresistant E. coli SER6) were used in ultraviolet disinfection experiments. The inactivation rates of E. coli strains under different ultraviolet irradiation doses were determined by plate count method. The impacts on antibiotic resistance and cell morphology were investigated by K-B disk diffusion method and scanning electron microscope,respectively. Under ultraviolet irradiation dose of 20 m J/cm2, the inactivation rates of Escherichia coli ATCC25922,SER2 and SER6 were 4. 75,5. 04 and 3. 83-log, respectively. The inactivation of E. coli ATCC25922 by ultraviolet disinfection was similar to that of E. coli SER2,while the inactivation rate of E. coli SER6 was lower than them. At a ultraviolet disinfection dose of 80 m J/cm2,the diameter of inhibition zone from gentamicin for E. coli SER6 changed significantly compared with that before disinfection( P〈0. 05). The structural integrity of bacteria may be destroyed by high ultraviolet irradiation dose(200 m J/cm2),which contributed to its inactivation.
