摘  要:目的:采用星点设计-效应面法优化柴胡-白芍药对超声提取工艺。方法:以乙醇浓度、料液比和提取温度为自变量,柴胡总皂苷和白芍总苷得率为因变量,通过对因变量的总归一值进行多元线性回归和二项式拟合,采用效应面优化最佳工艺条件。结果:确定最佳提取条件为:乙醇浓度77.60%,料液比23.24 m L/g,提取温度75℃,提取时间60 min,提取2次,二项式拟合的负相关系数r=0.9860,工艺验证的总评归一预测值与实测值的偏差为2.68%。结论:采用星点设计-效应面法优化柴胡-白芍药对的超声提取工艺,方法简单,预测性良好。Objective: To optimize the extraction process of total saponins from Bupleurum and Paeony by Central composite De- sign-Response surface Methodology. Methods : Ethanol concentration, extraction temperature and 'ratio of solvent to raw material were selected as independent variables. The overall desirability of the extraction rates of total saponins of Bupleurum and Paeony were set as dependent variables. Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables. Response surface methodology was used to optimize the process of extraction. Results: The optimal conditions of extraction process were determined as follows: Ethathol concentration 77. 60%, solvent-material ratio 23.24 mL/g, extraction temperature 75 ~C, extraction time 60 min and extract twice. The multiple correlation coefficient of binomial fitting of the complex model was as high as 0. 9860. The deviation between observed and predicted values was 2. 68%. Conclusions: The central composite design-re- sponse surface methodology is convenient and highly predictive for optimizing the extraction process of Bupleurum and Paeony.
