摘  要:建立了浊点萃取高效液相色谱法测定穿心莲中穿心莲内酯和脱水穿心莲内酯含量的方法。选择以表面活性剂Genapol X-080作为萃取剂,考察了表面活性剂浓度、液固比、盐浓度、平衡温度、平衡时间等因素对萃取结果的影响。获得最佳萃取条件为Genapol X-080的浓度为0.15 g/m L;液固比定为100∶1;盐浓度1.2 mol/L;平衡温度为60℃;平衡时间为30 min。浊点萃取是一种安全、高效、简便的样品处理方法,在最佳条件下,穿心莲内酯和脱水穿心莲内酯的加样回收率分别为95.4%和93.7%,相对标准偏差(RSD)分别为1.84%和2.31%。利用此方法成功测定了穿心莲中穿心莲内酯和脱水穿心莲内酯的含量。A new method was developed for the determination of andrographolide and dehydroandrographoiide content in Andrographis paniculata by cloud point extraction coupled with HPLC. The surfactant Genapol X -080 was chosen as the extract solvent. Some affecting factors such as the concentration of surfactant, liquid and solid ra- tio, concentration of sodium chloride, equilibration temperature and equilibration time were investigated and opti- mized. The optimum conditions were achieved of 0.15 g/mL surfactant concentration, 100:1 liquid and solid rati- o, 1.2 mol/L sodium chloride concentration for 30 rain under 60 tiC. This method is safe, efficient and easy. Un- der the optimum conditions, the average recoveries of andrographolide and dehydroandrographolide were 95.4% and 93.7% , while RSD were 1.84% and 2.31%. The method was successfully applied to determine androgra- pholide and dehydroandrographolide in Andrographis panieulata.
