摘  要:目的研究临床分离鲍曼不动杆菌生物被膜形成与外排泵基因表达的关系。方法平板培养结合结晶紫染色法建立鲍曼不动杆菌生物被膜模型,检测临床分离鲍曼不动杆菌的生物被膜形成能力,采用反转录聚合酶链反应(RT-PCR)和实时聚合酶链反应(Real-TimePCR)比较临床分离鲍曼不动杆菌在生物被膜形成的不同阶段外排泵基因adeB和adeJ的表达水平。结果随着生物被膜形成的成熟,外排泵基因adeB和adeJ的表达水平上调。结论临床分离鲍曼不动杆菌生物被膜形成与外排泵系统密切相关。Objective To study the correlation between biofilm formation and efflux pumps genes expression in clinical isolates of Acinetobacter baumannii. Methods Plates and crystal violet staining were used to construct a model of Acinetobacter baumannii biofilm formation in which the biofilm forming ability of clinical isolates was analyzed. Reverse transcription polymerase chain reaction and Real-time polymerase chain reaction were applied to detect the expression of the efflux pumps genes adeB and adeJ throughout the course of biofilm formation. Results Expression of the efflux pumps genes adeB and adeJ were upregulated throughout the course of biofilm formation. Conclusions Biofilm formation among the clinical isolates of Acinetobacter baumannii is closely correlated with efflux pumps.
