摘 要:为挖掘和应用海南疣粒野生稻优异资源,本研究调查和筛选优异疣粒野生稻资源,经穗颈注射法向R225等水稻恢复系导入疣粒野生稻基因组DNA,创新水稻育种中间材料;与三系、两系不育系测交配组,对F1代及后代观察、考种与测产,选育杂交水稻强优新组合。采用SSR标记检测导入创新种质、杂交水稻新组合与疣粒野生稻的遗传相关性。结果表明,筛选出15份优异疣粒野生稻资源,获得7份导入种质创新材料,与不育系测交配组选育出12份有效穗数、穗长、千粒重、抗病等农艺性状优良的杂交水稻新组合,通过品种比较试验,筛选出5份高产抗病水稻新组合。SSR标记检测导入创新种质、杂交新组合具有疣粒野生稻特有分子标记带型,说明获得来自疣粒野生稻的种质创新材料,并可稳定遗传。本研究为海南疣粒野生稻种质创新和进一步研究应用提供参考。To excavate and utilize germplasms resources of Oryza meyeriana Baill. indigenous to Hainan,the high resistance and vigorous growth of Oryza meyeriana Baill. indigenous to Hainan were investigated and screened out,then their genome DNA was extracted and transformed into rice restorer R225 by stem injecting. These innovation materials were testcrossed and combined with sterile lines,through observation of F1 and offsprings,agronomic traits test and varietal yield test the disease-resistant hybrid rice varieties of high yield and good quality were selected,and the correlation of these rice germplasm innovations and hybrid rice varieties with Oryza meyeriana Baill. were detected with SSR markers. The results showed that 15 accessions of high resistance and vigorous growth of Oryza meyeriana Baill. were screened out,7 rice variants of genome DNA transformation were found among those different filial plants,which were testcrossed with sterile lines to get 12 new rice varieties of excellent agronomic traits,and 5 new rice varieties of high resistance and high yield were chosen by comparative test.The results of SSR analysis indicated that there existed much polymorphism of repetitive DNA sequences between the variants and the receptors,with special DNA segments from distant relative Oryza meyeriana Baill.( donors) integrated into the genome of rice. The above mention would provide a reference for the research of Oryza meyeriana Baill. germplasm resources innovation and rice breeding.
