摘  要:目的研究唐古特大黄多糖对RAW264.7巨噬细胞免疫功能的调节作用。方法以小鼠腹腔巨噬细胞系RAW264.7为研究对象,分不同浓度的唐古特大黄多糖实验组、脂多糖阳性对照组、不加任何样品的空白对照组。通过MTT实验、吞噬中性红实验、一氧化氮释放量及细胞因子(IL-1β、IL-6、IFN-α、IFN-γ)的分泌水平,研究唐古特大黄多糖的免疫调节作用。结果实验表明唐古特大黄多糖在一定浓度范围可以促进吞噬细胞的增殖,并可增强其吞噬作用;唐古特大黄多糖可以有效提高巨噬细胞产生NO的量,且对巨噬细胞的细胞因子IL-1β、IL-6、IFN-α、IFN-γ的分泌量均有一定程度的提高。结论唐古特大黄多糖具有较好的提高巨噬细胞免疫功能的作用。Objective To investigate the influence of rheum tanguticum polysaccharides on the immunomodulatory effectsofRAW264.7macrophage. Methods To take murineintraperitonealRAW264.7macrophage as the research objects and different concentrations of rheum tanguticum polysaccharides and LPS injected into mice as positive control, without any treating asthe blank control. The influence of rheum tanguticum polysaccharides on the immunomodulatory effectsofRAW264.7macrophage was detected by MTT,phagocytosis test, NO release and the secretion of cytokines such as IL - 1β, IL - 6, IFN - α and IFN - γ. Results Experiments showed that rheum tanguticum polysaccharides can accelerate the proliferation of phagocytic cells to have a good phagocytosis. Besides, it can improve the macrophage to generate NO,and had a good promotion in enhancing the secretion of cytokines such as IL - 1β, IL - 6, IFN - α and IFN -γ Conclusion Rheum tanguticum polysaccharides can obviously enhance the function of macrophage.
