摘 要:目的:研究氧化苦参碱浓度、灌流速度及试验方法对同心圆和线性两种微透析探针回收率的影响。方法:以氧化苦参碱为试验药物,分别测定药物不同浓度、不同灌流速度下,两种探针体外正向、反向回收率及体内反向回收率。结果:探针回收率均与药物浓度无关,与灌流速度成反比;两种探针各自的体外正向、反向回收率相似,且均大于体内反向回收率;同心圆探针的回收率大于线性探针。结论:探针回收率的测定是采用微透析研究药动学的重要试验,可为氧化苦参碱的体内药物浓度确定指明适用的方法。Objective: To evaluate the effects of drug concentration, flow rate and method on the recoveries of two different types of concentric cannula and linear microdialysis probes. Methods: Oxymatrine was selected as model drug. In vitro recovery of oxymatrine was studied using positive dialysis and retrodialysis methods at different flow rates and concentrations. And in vivo recovery of oxymatrine was studied using retrodialysis methods. Results: The microdialysis recoveries of oxymatfine were inversely proportional to the perfusing rates, independent of oxymatrine concentrations. In vitro, the positive dialysis recoveries were similar to the retrodialysis recoveries. While the in vitro recoveries were greater than those in vivo. Under the same conditions, the recovery of concentric cannula probe was greater than that of linear probe. Conclusion: The determination of probe recovery is an important test for pharmacokinetics research on the basis of microdialysis, which indicates the appropriate and applicable way to measure the drug concentration of a certain kind of drug in the body.
