高血压病血瘀证血管内皮损伤的内质网应激细胞凋亡机制研究

摘　　要：目的:通过观察高血压病血瘀证患者血清致伤内皮细胞后与内质网应激蛋白JNK、CHOP相关的细胞凋亡,探讨高血压病血瘀证血管内皮损伤的机制。方法:运用免疫印记杂交技术检测各组内皮细胞内质网应激蛋白PERK、ATF6和IRE1的表达和内质网应激细胞凋亡相关蛋白JNK、CHOP的表达;以JNK和CHOP分别阻断与JNK、CHOP相关的内质网应激细胞凋亡通路,运用流式细胞术检测各组细胞的凋亡率。结果:PERK、ATF6和IRE1的表达,血瘀证组与正常组比较,差异均有统计学意义(P<0.01)。各组细胞凋亡率,与正常组比较,除血瘀证+CHOP、JNK阻断组外差异均有统计学意义(P<0.01);血瘀证+CHOP组与血瘀证组比较,差异有统计学意义(P<0.01);血瘀证+JNK组、血瘀证+CHOP组与血瘀证+JNK、CHOP组比较,差异有统计学意义(P<0.05,P<0.01)。各组JNK的表达,血瘀证组与正常组、非血瘀证组比较差异有统计学意义(P<0.01,P<0.05);血瘀证+JNK组、血瘀证+JNK、CHOP组与血瘀证组比较,差异有统计学意义(P<0.01)。各组CHOP的表达,血瘀证组与正常组、非血瘀证组比较,差异有统计学意义(P<0.01);血瘀证+JNK、CHOP组、血瘀证+CHOP组与血瘀证组比较,差异有统计学意义(P<0.01)。结论:高血压病血瘀证患者血清诱导人脐静脉内皮细胞发生了与内质网应激相关的细胞凋亡,与JNK、CHOP相关的内质网应激细胞凋亡导致的内皮细胞损伤,可能是高血压病血瘀证的重要发病机制。Objective： To study the mechanism of vascular endothelial injury induced by blood stasis syndrome associated with Essential Hypertension through observing the apoptosis about JNK and CHOP of vascular endothelial cell related to endoplasmic recticulum stress, which was based on the vascular endothelial injury model induced by blood stasis syndrome associated with Essential Hypertension. It was thought to be able to provide evidence for the prevention and treatment of Essential Hypertension. Methods： Detected the expression of the protein related to unfold protein response PERK, ATF6 and IRE1 in the blood stasis syndrome group, the non-blood stasis syndrome group and the normal group by western blotting method; Suppressed the JNK apoptosis signal pathway with its characteristic inhibitor SP600125; Inhibited the expression of CHOP with its inhibitor Doxorubicin, then observed the apoptosis rate of all experimental groups with SP600125 and Doxorubicin with flow cytometry; After SP600125 and Doxorubicin were added in for 24 hours, we measured the expression of JNK, CHOP relevant to endoplasmic recticulum stress apoptosis in all experimental groups with Western blot. Results： Compared to the normal group, the expression of PERK, ATF6 and IRE1 in the blood stasis syndrome group had significant difference （P〈0.01） after the model being established. Compared to the non-blood stasis syndrome group and the normal group, The expression of ATF6 in the normal group both had significant difference （P〈0.01）. Compared to the normal group, the late apoptosis rate in other groups except the blood stasis syndrome group with SP600125 and Doxorubicin had significant difference （P〈0.01）; the blood stasis syndrome group with Doxorubicin, the blood stasis syndrome group with SP600125 had significant difference （P〈0.01）; Compared to the blood stasis syndrome group with SP600125 and Doxorubicin, the blood stasis syndrome group with SP600125 had difference （P〈0.05）, the blood stasis syndrome group with Do
