摘  要:目的观察参附注射对脓毒症大鼠心肌细胞凋亡及Bcl-2蛋白表达的影响,探讨参附注射液抗脓毒症心肌损伤的相关机制。方法将24只雄性SD大鼠随机分为3组:假手术组、模型组和参附组,以盲肠结扎穿孔(CLP)法建立脓毒症大鼠动物模型,观察各组大鼠心肌酶谱变化;凋亡原位末端标记法(TUNEL)检测心肌细胞凋亡指数,以RT-PCR法检测心肌细胞Bcl-2 m RNA表达。结果参附组大鼠心肌酶谱升高程度较模型组低(P<0.05),模型组较假手术组心肌酶水平高(P<0.05);参附组大鼠心肌细胞TUNEL评分平均光密度(MOD)较模型组明显降低(P<0.05),模型组较假手术组升高(P<0.05);模型组心肌细胞Bcl-2 m RNA表达较假手术组明显降低(P<0.05),参附组较模型组升高(P<0.05)。结论参附注射液能减轻脓毒症大鼠的心肌损伤,其作用机制与上调Bcl-2 m RNA表达、减少心肌细胞凋亡相关。Objective: To investigate the anti-myocardial injury mechanism of Shengfu Injection by the change of myocardial enzyme and the expression of Bcl-2 and myocardial cell apoptosis in rat model with sepsis. Meth- ods: 24 male SD rats were randomly divided into three groups: the sham-operated group,the model group and the Shenfu group, 8 rats in each. Rat sepsis models were established with CLP. The change of myocardial enzyme spectrum was observed. Myocardial apoptosis was detected with TUNEL,and the expression of Bcl-2 mRNA in myocardial cell was detected with RT-PCR. Results: The rise of myocardial enzyme spectrum in Shengfu Injec- tion group was lower than that of the model group (P〈 0.05). Myocardial enzyme level of the model group was higher than that of the sham-operated group (P〈 0.05). TUNE L scores of cardiac muscle cells and MOD of Shen^u Injection group was lower than that of the model group (P〈 0.05), and higher than that of the sham-oper- ated group (P〈0.05). The expression level of Bcl-2 mRNA in myocardial cell of model group was lower than the sham-operated group (P〈 0.05),while the Shengfu Injection group was higher than the model grou (P〈 0.05). Conclusons: Shengfu Injection can alleviate injury in cardiac muscle caused by sepsis by upregulating the ex- pression level of BCL-2 and reducing myocardial apoptosis.
