摘  要:目的探讨褪黑素(MT)对丙烯酰胺(AA)诱发大鼠睾丸氧化损伤和细胞凋亡的作用。方法雄性大鼠随机分为对照组、AA染毒组和MT治疗组,每组各8只动物。实验结束后,取材各组大鼠的睾丸和血清,检测MDA和SOD指标,并利用免疫组织化学法检测睾丸组织中Bax和Bcl-2表达情况。结果与对照组相比,AA染毒组大鼠睾丸组织和血清中MDA含量明显增加(P<0.05);SOD活力显著下降(P<0.01),然而,经MT治疗后,大鼠睾丸和血清中MDA含量明显减少(P<0.05);SOD活力明显升高(P<0.05,P<0.01)。免疫组织化学的结果表明,与对照组相比,AA染毒组大鼠睾丸组织中Bax表达为强阳性,而Bcl-2表达呈弱阳性。经MT治疗后,可下调Bax的表达以及上调Bcl-2在睾丸组织中的表达。结论 MT能够有效缓解AA所致的大鼠睾丸毒性,且MT可能通过线粒体功能相关的抗氧化损伤和抗凋亡特性从而发挥调节作用。Objective To investigate the protective effects of melatonin on testicular oxidative damage and cell apoptosis induced by acrylamide in rats. Methods The male rats were randomly assigned to the control group, AA group, and AA + MT group.There were eight rats each group. After all rats were sacrificed, the testis and serums were sampled to detect their MDA and SOD levels. Immunohistochemical staining was applied to observe the changes of Bax and Bcl-2 levels in the testis. Results The MDA levels of the testis and serum in the AA group evidently increased in comparison with the control group(P〈0.05), while the SOD activities in the AA group were significantly reduced(P〈0.01). However, decreased MDA(P〈0.05) and elevated SOD(P〈0.05, P 0.01)was observed in testis and serum of AA + MT rats than these in the AA group. The immunohistochemical results showed that there was a higher expression of Bax in the AA group when compared to the control while the Bcl-2 positive expression was lower. MT treatment could modulate the downregulation of Bax expression and upregualtion of Bcl-2 expression in the testis compared with the AA rats. Conclusion The present results indicate that MT can performed an effectively protection against AA-induced testicular toxicity in rats. The protective effect of MT is mediated through anti-oxidative damage and anti-apoptotic properties associated with function of mitochondria.
