摘 要:目的:优选大孔树脂纯化展毛地椒总黄酮的工艺参数。方法:采用紫外分光光度法测定总黄酮含量,以总黄酮的吸附率和解吸率为指标,考察大孔树脂型号、上样液浓度、上样流速、洗脱剂种类及其用量等影响因素,优选最佳分离纯化条件。结果:AB-8树脂具有较好的吸附率和解吸率,最佳纯化工艺:上样液浓度为0.2 g/m L,上样流速为2 BV/h,上样体积为2 BV,洗脱溶剂为50%乙醇,洗脱体积为5 BV,径高比为1/6,富集物中展毛地椒总黄酮含量达到26.32%。结论:AB-8型大孔树脂能较好地用于分离纯化展毛地椒总黄酮。Objective:To optimize the purification technology parameters of total flavonoids from Thymus using macroporous resins. Methods:The content of total flavonoids in Thymus was determined by ultraviolet spectrophotometry spectrophotometer. With the adsorption and desorption capacity as index, purification technology was optimized by investigating the type of macro- porous resin, concentration of sample liquid ,flow speed for adsorption, type and amount of eluent solvent and other indicators. Re- suhs:AB -8 macroporous resin showed the good property for the absorption and desorption capacity for total flavonoids in Thy- mus. The optimized conditions were as follows : the concentration of sample liquid was 0.2 g/mL; the flow rate was 2 BV/h ; the e- lution solvent was 50% ethanol;the volume of elution solvent was 5 BV and the ratio of diameter to height was 1/6. Under the op- timized purification condition, the content of total flavonoids was up to 26. 32% in the enriched product. Conclusion:AB -8 macroporous resin can be effectively applied to purify total flavonoids from Thymus.
