摘 要:目的 检测瘦素对体外培养的不同免疫功能状态下的小胶质细胞株免疫功能的影响,探讨瘦素对BV2小胶质细胞株免疫调节作用.方法 体外模拟BV2小胶质细胞株炎性反应,分为空白对照组、脂多糖组(含2 mg/L脂多糖)、瘦素组(含2 mg/L瘦素)、脂多糖+瘦素组(含2 mg/L脂多糖和2 mg/L瘦素).药物干预24 h后,通过酶联免疫吸附试验法(ELISA)、流式细胞技术(FCM)、反转录-聚合酶链反应(RT-PCR)检测相关炎性因子的表达.结果 ELISA结果显示:瘦素组[(123.3 ±0.5)ng/L]较空白对照组[(114.6±1.1) ng/L]白细胞介素(IL)-1β分泌量升高,脂多糖+瘦素组[(134.0±0.5) ng/L]较脂多糖组[(142.4 ±0.3) ng/L]IL-1β分泌量降低,差异有统计学意义(P<0.05);FCM结果显示:瘦素组CD45、CD54的表达量(30.57±1.96、73.53 ±2.21)较空白对照组(21.64±2.58、61.22±2.81)升高,脂多糖+瘦素组(27.85±2.24、73.44±3.53)较脂多糖组(42.81±3.26、80.38±2.06) CD45、CD54的表达降低,差异有统计学意义(P<0.05),而细胞核增殖抗原(Ki-67)的表达在3组间差异无统计学意义(P>0.05);RT-PCR结果显示:瘦素组(10.55±1.31、7.96±0.57、0.87±0.13)较空白对照组(8.18±0.56、5.44±0.29、0.78±0.05)细胞凋亡蛋白因子配体(FasL)、诱导型一氧化氮合酶(iNOS)、肿瘤坏死因子-α(TNF-α)表达升高,脂多糖+瘦素组(10.64±1.24、8.08±0.46、0.91±0.11)较脂多糖组(20.55±2.59、9.51±0.69、1.23±0.12)FasL、iNOS、TNF-α表达有所降低,差异有统计学意义(P<0.05).结论 瘦素对BV2小胶质细胞株免疫功能具有双向调节作用:瘦素促进静息状态下BV2小胶质细胞株炎性反应;但对脂多糖诱导已活化的小胶质细胞株,瘦素则发挥抑制其炎性反应的作用.Objective Through detecting the effect of leptin on immune function of microglial cells cultivated in different conditions in vitro,this paper aims to study the immune adjustment of leptin on BV2 microglial cells.Methods Imulating BV2 microglial cell lines inflammatory reaction in vitro,Divided into blank control group,lipopolysaccharide group (Containing 2 mg/L lipopolysaccharide),leptin group (Containing 2 mg/L leptin),lipopolysaccharide + leptin (Containing 2 mg/L lipopolysaccharide and 2 mg/L leptin).After intervention for 24 h,Detecting the expression of related inflammatory factors using enzyme linked immunosorbent assay (ELISA)/flow cytometry (FCM)/reverse transcription-polymerase chain reaction (RT-PCR) technical methods.Results ELISA showed that the interleukin (IL)-1 β secretion in leptin group [(123.3 ± 0.5) ng/L] was significantly higher than blank control group [(114.6 ± 1.1) ng/L],while the IL-1 β secretion in lipopolysaccharide + leptin group [(134.0 ± 0.5) ng/L] was significantly lower than lipopolysaccharide group [(142.4 ±0.3) ng/L];the difference was statistically significant (P < 0.05).FCM showed that the expression of CD45 and CD54 in leptin group (30.57 ± 1.96,73.53 ± 2.21) was significantly higher than blank control group (21.64 ± 2.58,61.22 ± 2.81),while the expression of CD45 and CD54 in lipopolysaccharide + leptin group (27.85 ± 2.24,73.44 ± 3.53) was significantly lower than lipopolysaccharide group (42.81 ± 3.26,80.38 ± 2.06);the difference was statistically significant (P < 0.05).There was no statistical difference between three groups on the expression of Ki-67.RT-PCR showed that the expression of FasL,inducible nitric oxide synthase (iNOS) and tumor necrosis factor (TNF)-α in leptin group (10.55 ± 1.31,7.96 ± 0.57,0.87 ±0.13) was significantly higher than blank control group (8.18 ±0.56,5.44 ±0.29,0.78 ±0.05),while the expression of FasL,iNOS and TNF-α in lipopolysacch
