摘  要:利用已报道的SRK及SLG基因序列保守区设计的特异性引物,通过PCR扩增和克隆测序,结合生物信息学分析方法对4份国外引进的不结球白菜(Brassica campestris ssp.chinensis Makino)自交不亲和材料的S单元型进行分子鉴定。结果表明,不亲和材料A1包含的S位点基因序列与甘蓝(B.oleracea L.var.capitata L.)SLG-31的相似度为98%(E值是0);不亲和材料A2、A3包含Class I类及ClassⅡ类2种S单元型,S位点处于杂合状态。不亲和材料A4包含的S位点基因序列与青花菜(B.oleracea L.var.italica Plenck)单倍型BOI1 SRK蛋白基因具有98%的序列相似度,序列覆盖度达99%。Using the primers designed according to the conserved sequence of S locus kinase(SRK) gene and S locus glycoprotein(SLG) gene, by PCR amplification, combined with bio-informatics analysis the S haplotypes of 4 Brassica campestris ssp. chinensis Makino self-incompatibility material from overseas were identified. The results indicated that the A1self-incompatible line showed 98% similarity with SLG-31 gene in B. oleracea L. var. capitata L.; and the A2, A3 self-incompatible line both contained two types of S haplotype from Class I and Class Ⅱ, which showed that the S locus were at heterozygous state. The A4 self-incompatible line showed 98% similarity with BOI1 SRK gene in B. oleracea L. var. italica Plenck under 99% query coverage.
