摘  要:制备高效价的幽门螺杆菌尿素酶B(Ure B)亚单位重组蛋白免疫血清,探讨不同剂量、不同途径对家兔免疫效果的影响。采用0.5,1,5 mg/m L Ure B抗原分别通过皮内(sc)、肌肉(im)、腹腔(ip)3种途径对新西兰大白兔进行免疫,免疫双扩散法测定抗血清效价。结果显示,1 mg/m L抗原剂量免疫效果最佳,优于0.5 mg/m L的剂量,5 mg/m L的抗原剂量造成家兔免疫耐受;皮内注射途径所制备的兔抗血清抗体效价最高。本研究成功制备Ure B重组蛋白多抗血清,同时还优化得到Ure B重组蛋白多抗血清制备的最佳免疫剂量和免疫途径,为今后幽门螺杆菌疫苗的活性测定奠定了基础。To prepare effective immune serum of Helicobacter pylori urease B subunit recombinant protein and to discuss the immune effect in different doses and by different routes,the highly purified Helicobacter pylori urease B subunit recombinant protein was used to immune rabbit through three different doses and by three different routes. The antiserum titers were evaluated by double immunodiffusion in agar gel. The dose of 1 mg / m L could present better effect than the dose of 0. 5mg / m L in this research,while the dose of5 mg / m L caused immune tolerance of the rabbits. The antibody obtained from the intradermal administration showed the higher lever than that from the intrmuscular or the intraperitoneal administration. The research successfully obtained effective immune serum of the Helicobacter pylori urease B subunit recombinant protein and further expolred the antiserum preparation protocols including the optimal immune dose and immune route,which could establish a foundation for the research and development of Helicobacter pylori urease B subunit recombinant vaccine in the future.
