摘 要:研究牛磺胆酸(taurocholic acid,TCA)对NR8383细胞分泌的肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的影响。采用实时荧光定量PCR(real-time quantitative,qPCR)法测定TCA对细胞外信号调节激酶(extracellular signal-regulated protein kinase,ERK1/2)抑制剂(PD98059)介导的脂多糖(lipopolysaccharides,LPS)诱导的TNF-αmRNA水平的影响,采用酶联免疫吸附法(enzyme-linked immune-sorbent assay,ELISA)测定TCA对PD98059介导的NR8383细胞分泌的TNF-α蛋白水平相对表达量的影响。TCA可显著降低LPS诱导的TNF-α基因、蛋白水平含量(P<0.05),PD98059可显著降低TNF-α的表达量。TCA通过影响ERK1/2通路调节LPS诱导的NR8383细胞TNF-α基因、蛋白水平含量。It is important to study effects of taurocholic acids (TCA,a kind of natural bioactive sub- stance of animal bile acids) on the production of TNF-α in NR8383 cells for its comprehensive pharmacological effects. Intracellular TNF-α mRNA production in cells was measured by real-time quantitative(qPCR), Intracellular TNF-α protein production in cells was measured by enzyme- linked immune-sorbent assay. TCA decreased the production of TNF-α in cells induced by PD98059;TCA decreased the production of TNF-α mediated by LPS in NR8383 cells, too. TCA de- creased the production of TNF-α induced by LPS cooperated with PD98059 in NR8383 cells. TCA decreased the production of TNF-α induced by LPS associated with ERK1/2 signaling pathway in NR8383 cells.
