摘  要:观察雌激素(β-雌二醇)对奶牛输卵管上皮细胞膜结合型前列腺素E2合酶-1(mPGES-1)表达的影响,并探讨β-雌二醇对奶牛输卵管上皮细胞合成分泌PGs调控的作用机制。采用胰酶消化法及机械法分离培养奶牛输卵管上皮细胞,应用荧光定量RT-PCR技术检测β-雌二醇对奶牛输卵管上皮细胞mPGES-1 mRNA表达的影响,应用Incell Western技术检测β-雌二醇对奶牛输卵管上皮细胞mPGES-1蛋白表达的影响。结果显示,与空白对照相比,β-雌二醇能够显著和极显著地促进奶牛输卵管上皮细胞中mPGES-1基因和蛋白的表达。本试验结果表明,β-雌二醇能够对奶牛输卵管上皮细胞中mPGES-1产生调节作用。The study aimed at observing effect of β-estradiol on mPGES-1 expression and investi- gate the mechanism on the regulation of β-estradiol on the synthesis and secretion of PGs in bovine oviduct epithelial cell. Bovine oviduct epithelial cells(BOECs) were isolated and cultured by me- chanical and pancreatic enzyme digestion method;mRNA expression of mPGES-1 was detected by real-time RT-PCR in bovine oviduct epithelial cells;protein production of mPGES-1 was detected by Incell Western. The results show that compared with the blank control,β-estradiol markedly promoted the mRNA and protein expression levels of mPGES-1 in bovine oviduct epithelial cells. It is concluded that β-estradiol can regulate the expression levels of mPGES-1 in bovine oviduct epi- thelial cells.
