摘  要:目的研究JNK在Na As O2诱导L-02肝细胞凋亡中的作用。方法用不同浓度Na As O2染毒L-02肝细胞,采用流式细胞术检测L-02肝细胞凋亡率;Western-blot检测L-02肝细胞中Jnk、p-Jnk、Caspase-3蛋白的相对表达量。结果 0、50、100和150μmol/L组细胞凋亡率分别为3.33%±0.30%、7.49%±0.23%、9.48%±0.49%和14.87%±2.17%,染毒组的细胞凋亡率随Na As O2浓度的增加而升高,差异有统计学意义(P<0.05);染毒组细胞内Jnk、p-Jnk、Caspase-3蛋白相对表达量均增高,差异有统计学意义(P<0.05)。结论 Na As O2能诱导L-02肝细胞凋亡的发生,这可能与激活JNK,增加JNK磷酸化水平,继而激活凋亡调控基因Caspase-3有关。Objective To study the role of JNK signaling pathways in L-02 liver cell apoptosis induced by NaAsO2. Methods We infected L-02 liver ceils with different concentrations NaAsO2, and detected the apoptosis rate of L-02 liver cells by the flow cytometry and the relative expression quantities of JNK, p-JNK and Caspase-3 proteins in L-02 liver cells by the western-blot. Results The apoptosis rates of the groups with the concentrations of 0, 50, 100 and 150 μmol/L were 3.33% ± 0.30%, 7.49% ±0.23%, 9.48% ± 0.49% and 14.87% ± 2.17%, respectively. The apoptosis rate of infected group raised with the increasing concentration of NaAsO2, and the difference was statistically significant (P〈0.05). The relative expression quantities of JNK, p-JNK and Caspase-3 proteins in the infected group all increased, and the difference was statistically significant (P〈0.05). Conclusion NaAsO2 can induce the apoptosis of L-02 liver ceils. It may be related to the activation of JNK, increasing level of p-JNK and activation of Caspase-3 related gene.
