摘　　要：目的评价外用0.5%5-氟脲嘧啶(5-Fu)治疗日光性角化病(AK)的疗效及安全性。方法计算机检索Cochrane图书馆、Medline,Embase,NGC和CBM,纳入所有0.5%5-Fu治疗AK的随机对照试验。由2名研究者独立进行质量评估和交叉核对,并采用RevMan 4.3软件进行统计分析。结果共纳入3篇随机对照试验(RCT),包括528例患者,meta分析结果显示:疗程1周、2周及4周临床疗效均优于安慰剂,差异有统计学意义(P<0.05);疗程4周的临床疗效优于疗程1周及2周,差异有统计学意义(P<0.05)。不良反应主要为局部皮肤刺激,随疗程的延长而增加,但多为轻中度,患者具有较好的耐受性。结论 0.5%5-氟脲嘧啶治疗日光性角化病是安全有效的。Objective To assess the effect and safety of 0. 5% 5 - flurouracil in the treatment of Actinic keratosis. Methods The Cochrane library,Medline,Embase,NGC and CBM were searched,the randomized controlled trials（RCTs）about 0. 5%5 - flurouracil for the treatment of actinic keratosis all over the world were included. Two reviewers independently screened the studies for eligibility,evaluated the quality with confirmation by cross - checking. The data were inputted and analyzed by Rev-Man 4. 3 software. Results Three RCT studies involving 528 patients met the inclusion criteria. Meta analysis of the treatment of 0. 5% 5 - flurouracil showed treatment applied once daily for 1,2,or 4 weeks was more effective than vehicle control in lesion clearance. And the treatment for 4 weeks was better than one week and two weeks. Commonly reported adverse events were local-ized facial irritation. Most of them were mild or moderate,and most of these patients were able to tolerate it. Conclusion Topical 0. 5% 5 - flurouracil was an effective and safe regimen for the treatment of actinic keratosis.
