健心颗粒对慢性心衰大鼠心肌病理及心功能的影响

摘　　要：目的探讨健心颗粒对慢性心衰大鼠心肌病理及心功能的影响。方法将15周龄的自发性高血压大鼠(SHR-sp)30只随机分为培哚普利组、健心颗粒低剂量组、健心颗粒中剂量组、健心颗粒高剂量组、阴性对照组,每组6只。前四组分别予培哚普利0.36mg·(kg·d)-1、健心颗粒1.6 g·(kg·d)-1、3.2 g·(kg·d)-1、6.1 g·(kg·d)-1,以2ml生理盐水溶解,阴性对照组予生理盐水2ml,灌胃给药,每天2次。治疗前及治疗28天后断尾取血,采用ELISA法检测大鼠血浆B型钠尿肽(BNP),并处死后取心室肌组织,行HE染色,光学显微镜下观察组织形态学变化。结果低、中、高剂量健心颗粒及培哚普利组能显著降低慢性心衰大鼠BNP水平,病理显示心室肌炎细胞浸润减少。结论健心颗粒能显著改善慢性心衰大鼠心肌病理及心功能。Objective To investigate the effect of the Jianxin granule （ JG ） on cardiac function and myocardial pathology of rats with chronic heart failure.Methods 30 cases of 15 weeks of spontaneously hypertensive rats （ SHR-sp） were randomly divided into five groups：Perindopril group 0.36mg·（kg·d）^ -1, low dose JG group 1.6g·（kg·d） ^-1, median dose JG group 3.2g·（kg·d）^ -1, high dose JG group 6.1g·（kg·d） ^-1 and negative control group （ with saline 2 ml） , and each group had 6 cases.The drugs were administrated by ig for 2 times a day, dissolved in 2 ml saline water.Before and after 28 days’ treatment, blood samples were collected by cutting rats’ tails.We detected plasma BNP of rats by using the method of ELISA.After rats were executed, the ventricular muscle tissue was examined though HE staining, and changes of histomorphology were observed under the optical microscope.Result Low, medium and high dose JG groups and Perindopril group can significantly reduce the BNP levels of chronic heart failure rats, and pathology revealed that abnormal structure of the myocardial cell reduced. Conclusion Jiaxin granule could significantly improve myocardial pathology and cardiac function of rats with chronic heart failure.
