摘 要:目的观察溪黄草水提物对α-萘异硫氰酸酯(ANIT)诱发的肝内胆汁淤积大鼠的影响。方法采用ANIT 80 mg·kg-1复制大鼠肝内胆汁淤积模型,健康SD雄性大鼠按体质量随机分为6组,每组8只,即:空白对照组,溪黄草高剂量组(60 g·kg-1),溪黄草中剂量组(40 g·kg-1),溪黄草低剂量组(20 g·kg-1),模型组和熊去氧胆酸(UCUD)组(100 mg·kg-1)。灌胃给药后,进行胆总管插管手术,观察溪黄草对大鼠1.5 h内的胆汁流量以及血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(γ-GT)、总胆固醇(TC)、直接胆红素(DBIL)、总胆红素(TBIL)、总胆汁酸(TBA)的影响。结果与模型组比较,溪黄草各剂量组各项指标(除ALT、ALP、γ-GT外)均有一定改善(P<0.05,P<0.01),但部分未呈现一定量效关系。UCUD组对AST有改善,其他7项指标效果不明显。与模型组比较,溪黄草中、高剂量组以及UCUD组胆汁流量均有显著提高(P<0.05,P<0.01)。病理切片结果显示溪黄草高剂量组和UCUD组对ANIT诱发的肝细胞损伤有所改善。结论溪黄草对ANIT诱发的实验性大鼠肝内胆汁淤积损伤具有一定干预改善作用。Objective To study the protective effect of Herba Rabdosiae Serriae(Xihuangcao) aqueous extract on rats with acute intrahepatic cholestasis induced by alpha naphthyl isothiocyanate (ANIT). Methods Animal models of intrahepatic cholestasis was established with ANIT at 80 mg·kg-1. Forty-eight Sprague Dawley rats were randomly divided into 6 experimental groups, namely normal control group, model group, and high-, middle- and low-dosage Xihuangcao groups(60, 40, 20 g·kg-1 respectively), ursodeoxycholic acid(UCUD) group (100 mg·kg-1), 8 rats in each group. After gavage administration, the rats were given common bile duct surgery and then we observed the bile flow within 1.5 h in rats and measured serum alanine transaminase(ALT), aspartate transaminase(AST), alkaline phosphatase(ALP), γ- glutamyl transferase(γ-GT), total cholesterol(TC), direct bilirubine(DBIL), total bilirubin(TBIL), and total bile aeid(TBA)levels in serum. Results Compared with the model group, high-, middle- and low-dosage Xihuangcao significantly decreased all of the observation indexes except ALT, ALP and γ-GT (P 〈 0.05), and the changes of some indexes were in dose-dependent manner. UCUD only had obvious effect on improving AST. Middle- and high-dose Xihuangcao group and UCUD group had higher bile flow than the model group(P 〈 0.01 ). The results of pathological examination showed that high-dose Xihuangcao and UCUD significantly relieved the injury of hepatocytes induced by ANIT (P 〈 0.01). Conclusion Xihtumgcao aqueous extract shows certain effect on improving the injury in rat with ANIT-induced intrahepatic cholestasis.
