摘 要:目的观察加减驻景方对病理性近视脉络膜新生血管(choroidal neovascularization,CNV)的血管内皮生长因子(vascular endothelial growth factor,VEGF)及色素上皮衍生因子(pigment epithelium-derived factor,PEDF)表达的影响,探讨中药驻景方对病理性近视CNV的干预作用。方法将3周龄雌性三色豚鼠75只随机选取15只为空白对照组,剩余豚鼠均戴头套右眼形觉剥夺诱导病理性近视,并随机分为模型组、中药组,每组各30只,右眼行氪激光光凝。于光凝后第1天开始连续灌胃21 d,中药组予驻景方3.285 g·kg-1·d-1(1.5 m L·d-1),模型组予等量的生理盐水。造模后7 d、14 d、21 d分别行免疫组织化学及RTPCR检测。结果免疫组织化学结果显示,造模完成后14 d、21 d时中药组VEGF表达的光密度值(0.130 1±0.019 6、0.055 6±0.011 9)较模型组(0.165 9±0.054 3、0.096 1±0.025 9)减少,差异均有统计学意义(P=0.006,P<0.001)。造模完成后14d及21 d时中药组PEDF表达的光密度值(0.038 9±0.004 0、0.054 3±0.008 5)较模型组(0.030 5±0.007 7、0.029 3±0.0059)增高,差异有统计学意义(P=0.026,P<0.001)。RT-PCR结果显示,VEGF mRNA、PEDF mRNA在各组中均有表达,中药组VEGF mRNA表达水平较模型组明显减弱,而PEDF mRNA的表达较模型组增强。结论中药驻景方可抑制氪激光诱导的病理性近视CNV模型中VEGF的表达,促进PEDF的表达,从而抑制CNV的形成。Objective To observe the effects of modified Zhu Jing prescriptions on the expression of vascular endothelial growth factor (VEGF) and pigment epitheli- urn-derived factor (PEDF) in pathologic myopia with choroidal n (CNV) animal model,and explore the intervention effects of it on the pathological myo- pia with CNV. Methmll Fifteen form seventy-five 3-week old female guinea pigs were randomly selected as the blank control group. The rest 50 guinea pigs were used face- mask to induce pathologic myopia, then were randomly assigned to model group and treatment group, 30 cases in each group, the right eyes were performed the krypton la- sers photocoagulation. The treatment group was gavaged with modified Zhu Jing pre- scriptions 3. 285 g·kg-1· d-1 ( 1.5 mL·d-1 ) for 21 days,while the control group was gavaged with distill water 1.5 mL·d-1. Immunohistochemistry and RT-PCR were per- formed to detect the expression of VEGF and PEDF at 7 days, 14 days and 21 days after modeling. Resaults The immunohistochemistry showed that the AOD of VEGF expres- sion at 14 days and 21 days in the treatment group(0. 130 1 ±0.019 5,0.055 5±0.011 9) were less than those in the model group (0. 165 9 ± 0. 054 3,0. 095 1 ± 0. 025 9 ) ( P = 0. 05,P 〈 0. 001 ). AOD of PEDF expression at 14 days and 21 days in the treatment group(0.038 9±0.004 0,0.054 3 ± 0.008 5 ) were higher than those in the model group ( 0.030 5 ± 0.007 7,0.029 3±0.005 9 ) ( P = 0.026, P 〈 0.001 ). RT-PCR also showed the VEGF mRNA expression in the treatment group was less than that in the model group, and the PEDF mRNA expression in the treatment group was higher than that in the model group. Conclusion Modified Zhu Jing prescriptions can reduce VEGF expression and promote PEDF expression in pathological myopia with CNV. Thus, it may inhibit the formation of CNV.
