马钱子散中马钱子总生物碱的含量测定方法研究

摘　　要：目的：建立马钱子散中马钱子总生物碱的含量测定方法。方法：采用酸性染料比色法，溴百里香酚蓝为酸性染料，分别以马钱子碱和士的宁为对照品，测定马钱子散中马钱子总生物碱含量。结果：士的宁、马钱子碱在0．0200～0．0802mg／mL浓度范围内与吸光度均呈良好的线性关系，士的宁、马钱子碱的回收率分别为：95．30％-101．48％（RSD为2．28％），97．14％～105．30％（（RSD为2．95％）；以马钱子碱计，马钱子散中马钱子总生物碱含量为3．16％。结论：所建立的总生物碱测定方法准确、稳定、重复性好，可用于马钱子散中马钱子总生物碱的含量测定。Objective： To establish a method for quantitative determination of total alkaloids from seeds of Strychnos nux -vomica L. Methods： The content of total alkaloids in the samples was determined by the acid dye colorimetry ,which was performed with brucine and strychnine as the reference substances,bromine thymol blue as the acid dye and chloroform was used as the extractant. Result： Both the concentration of strychnine being 0. 0200 - 0.0802 mg/mL and that of brucine being 0.0200 - 0. 0802 mg/mL had a good linear relationship with the absorbance. According to the standard curve calculation of strychnine, the average content of total alkaloids was 2.54% and according to the standard curve cal- culation of brucine,the average content of total alkaloids was 3.16%. Conclusion： The established method is accurate and stable and has good repeatability and can be used for the determination of the content of the total alkaloids extracted from the seeds of Strychnos nux - vomica L.
