摘 要:目的:制定复方茸血胶囊中鹿茸多肽含量测定方法及制备工艺的研究。方法:采用正交实验设计。以出膏率为指标,优选杜仲、枸杞、当归、熟地黄提取最佳工艺中的溶媒用量、提取时间及提取次数。采用福林酚法测定复方茸血胶囊中鹿茸多肽含量。结果:最佳提取工艺第一次加水十倍量,煎煮2.0h,第二次煎煮时间1.5h,加水8倍量。鹿茸多耿线性范围:0.2142~0.4998μg。样品含量测定数据:不得少于36.8mg。结论:本法简便、准确,可用于复方茸血胶囊的含量测定。经过优化后制备的复方茸血胶囊各项指标符合规定,工艺重复性好。适于规模化生产。Objective:To establish the determination method for pilose antler polypeptide in compound antler blood capsule and preparation process of the capsule. Methods : The optimum extraction process of Eucommia ulmoides , Chinese wolfberry , Chinses Angelica and Radix Rehmanniae Preparata was determined by orthogonal experiment with the rate of paste as an index, different amounts of solvent, extraction time and extraction times as investigation factors. The component of pilose antler polypeptide in compound antler blood capsule was detected by Folin - Phenol reagent. Results : The optimum extraction process was the first time with ten times amount of water decocting for 2 hours, and the second with 8 times amount of water decocting for 1.5 hours. The linear range of pilose antler polypeptides : 0. 214 2 - 0. 499 8 μg. Content determination of sample data can not be less than 36.8 mg. Conclusion : The method is simple and accurate and can be used for determination of the content of compound antler blood capsule. After optimization of the compound antler blood capsule, indicators prepared are in accordance with the regulations and have good technique repeatability, suitable for large - scale production.
