摘  要:目的:探讨氢气对重度子痫前期(PE)胎盘滋养细胞的增殖和凋亡的影响。方法:MTT法检测不同浓度含氢培养基作用后滋养细胞的增殖活性。采用流式细胞术检测氢气作用24h后细胞凋亡率及细胞周期分布。结果:氢气能显著增强重度PE滋养细胞的增殖活性,抑制细胞凋亡(P<0.05),并使细胞周期中G1期比例减少,S期比例升高(P<0.05)。结论:氢气能显著提高重度PE胎盘滋养细胞的增殖活性,并能抑制细胞凋亡,促进细胞分裂。提示氢气在干预PE方面,可能是一种较为有效的抗氧化剂,具有潜在的预防和治疗价值。Objective:tion and apotosis to severe preeclampsia trophoblast cells. Methods:MTT was used to detect cell proliferation after trophoblast cells cultured by hrdrogen-rich medium. Flow cytometry was used to evaluate cell apotosis and cell cycle after treated by hydrogen-rich medium for 24h. Re-sults:Cell viability was significantly increased by 50 ~ 200μmol/ L molecular hydrogen ( P 〈0. 05). Meantime molecular hydrogen can inhibit cell apotosis (P〈0. 05),decrease the portion of G1 phrase cells (P〈0. 05) and raise the portion of S phrase cells (P〈0. 05). Conclusion:Molecular hydrogen significantly increases the proliferation of severe preeclampsia trophoblast cells in vitro,inhibits cell apotosis and blocks the cell cycle at S phrase. It suggests that H2 acts as an effective anti-oxidant,may possess the potential for preventive and therapeutic applications in PE.
