摘  要:为了探讨土壤线虫在人工林生态系统中的功能及其对环境变化的响应。该研究利用18S r DNA的宏基因组比较了湖南省攸县不同发育阶段(3年、13年、26年)以及不同栽培代数(1代、2代、3代)杉木人工林的土壤线虫的群落结构及多样性。结果表明:(1)测序共获得了2 111条隶属于线虫动物门的序列。经聚类后产生了27个OTUs。主要类群有Prismatolaimidae、Monhysteridae、Alaimidae、Qudsianematidae、Criconematidae、Plectidae它们分别占总序列数的31%、15%、11%、8%、6%、5%,其中Prismatolaimidae在所有样地中均占绝对优势,Aphelenchidae、Cephalobidae、Criconematidae、Rhabdolaimidae、Tylenchidae这5个类群的相对丰度在各样地间具有显著性差异(p<0.05);(2)冗余分析(RDA)发现所选的5块样地的线虫的群落结构有着明显的差异,FRC,SYC,TYC三块样地的群落结构较为相似。FYC与FRC之间差异性最大;(3)6种不同的多样性指数的单因素方差分析发现,各样地的多样性指数之间没有达到显著性差异。说明长期种植单一杉木人工林能够改变土壤线虫的群落组成,但并没有改变种群的多样性。土壤线虫中存在种的冗余现象。In order to study the function of soil nematode and its responses to environmental changes, we used metagenome analyses of 18S rDNA gene region to identify differences in nematode community structure and diversity among fifteen soil samples belonging to five different Cunninghamia lanceolate plantations. The plantations were located in Youxian County, Hunan Province in central China. The trees in these plantations were of different ages (3, 13, and 26 years) and belonged to different ecological successions (first, second, and third successions).We received a total of 2111 sequences which belonging to the phylum of nematode, Generated 27 OTUs after clustering.The dominant taxonomic groups across all samples were Prismatolaimidae, Monhysteridae, Alaimidae, Qudsianematidae, Criconematidae and Plectidae with each accounting for 31%, 15%, 11%, 8%, 6%, and 5%, and the group of Prismatolaimidae was dominant in all samples.The relative abundance of these five groups (Aphelenehidae, Cephalobidae, Criconematidae, Rhabdolaimidae and Tylenehidae) have significant difference (p 〈 0.05) among the five plantations. The redundancy analysis (RDA) identified that the community structure of nematode among the five plantations were quite difference, and it was similar among the first successions with the age of 26 years, the second successions with the age of 3 years and the third successions with the age of 3 years. The largest difference occurs between the first successions with the age of 3 years and the first successions with the age of 26 years. The ANOVA analysis with 6 different diversity indices (Shannon Index, McIntosh's Index, Simpson's Index, Dominance Index, Evenness Index and Berger-Parker Index) show that there were no significant difference among different types of Cunninghamia laneeolata plantations.This study indicated that long term single with pure plantations could affect soil nematode community, but it did not change the diversity of the population.
