摘 要:目的:探讨不同强度运动结合CLA对青春期肥胖大鼠Vaspin的影响。方法:建模7周后,取40只高脂诱导的肥胖大鼠随机分为5组,每组8只,分为对照组(OC)、静止+CLA组(OCC)、低强度运动+CLA组(OLC)、中强度运动+CLA组(OMC)、高强度运动+CLA组(OHC)。OLC、OMC、OHC分别以15~18 m/min、21~25 m/min和28~32 m/min的强度运动,坡度均为0°,1 h/d。每日运动后灌喂CLA 1.6 g/kg,5次/周,干预8周。8周后,采用qRT-PCR法检测脂肪组织Vaspin mRNA表达,采用免疫组化法检测脂肪细胞Vaspin蛋白,采用ELISA法检测血浆Vaspin浓度。结果:干预后OMC组、OHC组体重低于OCC组、OC组(P〈0.05或0.01),OLC组体重低于OC组(P〈0.05)。OLC组、OMC组、OHC组内脏脂肪重量和体脂率均低于OC组、OCC组(P〈0.01)。Vaspin mRNA:OLC组、OMC组高于OC组、OCC组(P〈0.01),OHC组高于OCC组(P〈0.05);OLC组、OMC组高于OHC组(P〈0.01)。Vaspin蛋白:OCC组蛋白表达最弱,OC组较OCC组蛋白表达明显,运动结合CLA相关组蛋白表达较OC组、OCC组强烈,且随着强度的增大,表达越明显。5组血浆Vaspin浓度差异均无统计学意义(P〉0.05)。结论:不同强度运动结合CLA干预8周后,较好地降低了青春期肥胖大鼠体重、内脏脂肪重量和体脂率,运动强度越大越明显;显著上调了Vaspin mRNA、Vaspin蛋白的表达水平,运动强度越大,上调越明显。Objective: To investigate the effect of different intensity exercise and conjugated linoleic acid (CLA) on Vaspin. Methods: High fat fed rat model was established after 7 weeks, 40 obesity rats were randomly divided into 5 groups, 8 rats in each group, respectively, the control group (OC), CLA group (OCC), low intensity exercise with CLA group (OLC), moderate in- tensity exercise with CLA group (OMC), high intensity exercise with CLA group (OHC). OLC, OMC, OHC in animal treadmill running speed respectively for 15-18 m/min, 21-25 m/min, 28-32 m/min, at an incline of 0 degrees, 1 h/day. For CLA treat- ment, each rat was fed 1.6 g/kg, 5 times/week, for 8 consecutive weeks. Blood and visceral adipose tissue of rats was taken after 8 weeks, qRT-PCR was used to measure the expression of Vaspin mRNA in adipose tissue, immunohistochemistry was used to measure the expression of Vaspin protein in fat cells, plasma Vaspin concentration was measured by ELISA. Results: After 8 week interven- tion, body weight in OMC group, OHC group were significantly lower than that in OCC group and OC group (P〈0.05 or 0. O1 ), body weight in OLC group was lower than that in OC group (P〈0.05). Visceral fat weight and FAT% in OLC, OMC, OHC groups were significantly lower than those in OC, OCC groups (P〈O. 01 ). Vaspin mRNA in OLC, OMC groups was significantly higher than that in OC, OCC groups (P〈O. 01), Vaspin mRNA in OLC, OMC groups was higher than that in OHC group (P〈0. 01), and Vaspin mRNA in OHC group was higher than that in OCC group (P〈0. 05). The expression of Vaspin protein in OCC group was weakest, the expression of Vaspin protein in OC group was more obvious than that in OCC group. The expression of Vaspin protein in the group of exercise with CLA was most obviously, and exercise intensity bigger and more obvious.
