摘 要:目的明确大鼠神经系统是否存在SIRT1-△Exon7,并初步探讨SIRT1-FL和SIRT1-△Exon7对神经干细胞分化的影响。方法分别提取新生大鼠海马和皮层的mRNA,应用PCR和RT-PCR方法检测SIRT1-FL和SIRT1-△Exon7的mRNA表达水平。结果 SIRT1-△Exon7和SIRT1-FL的mRNA在大鼠海马和皮层均有表达。皮层SIRT1-FL mRNA含量比海马高5.81%(P<0.05),皮层SIRT1-△Exon7 mRNA含量比海马低56.68%(P<0.05)。结论首次发现大鼠神经系统存在SIRT1-△Exon7。SIRT1-FL可能促进了神经干细胞的分化过程,而SIRT1-△Exon7可能发挥相反的作用。Objective To explore the expression of SIRT1- A Exon7 in rat nervous system and the effect of SIRT1-FL and SIRT1 alter- native splicing variants on neural stem cell differentiation. Methods The total RNA of newborn SD rat hippocampus and cortex was extracted, and the mRNA expression levels of SIRT1-FL and SIRT1-△Exon7 were detected in hippocampus and cortex by PCR and RT-PCR, respectively. Results The SIRT1- △ Exon7 and SIRT1-FL mRNA was expressed in both hippocampus and cortex of rats. The mRNA levels of SIRT1-FL was 5.81% higher in the cortex than that in the hippocampus (P 〈 0.05 ), whereas the S1RT1-△ Exon7 was 56.68 % lower in the cortex than that in the hippocampus ( P 〈 0.05 ). Conclusion The results firstly indicate that SIRT1- △ Ex- on7 exists in the nervous system of rats. SIRT1-FL may promote the differentiation of neural stem cells, while the SIRT1- △ Exon7 may play an opposite function in neural stem cells.
