摘 要:以L-脯氨酸为原料经过氨基保护、格氏试剂反应、脱保护合成了α,α-二苯基-2-吡咯烷甲醇,对影响反应的重要因素进行了考察和讨论。氨基保护时L-脯氨酸和氯苄物质的量最佳比为1∶1.5、反应温度为100℃。格氏试剂与N-苄基-L-脯氨酸苄酯的最佳物质的量比为4∶1。可以得到纯度为99%以上,收率为76.8%的产物。该合成方法简单、收率高,适合于放大生产。The α,α-diphenyl-2-pyrrolidinylmethanol was synthesized from L-proline via the processes of the amino protection, reaction with Grignard reagent and deprotection. The key factors which had effects on the reaction were investigated and discussed. In the process of amino protection, the optimum molecular ratio of L-proline to benzyl chloride was 1:1.5 and the reaction temperature was 100℃.The optimum molecular ratio of Grlgnard reagent to N-benzyl-L-proline benzyl ester was 4:1. The end product was synthesized with a purity of 99% and a yield of 76.8% under the conditions mentioned above. This method was simple with a high yield, which was suitable for the pilot scale.
