瑞舒伐他汀钙对兔血管平滑肌细胞TGF-β_1表达的影响

摘  要:目的 研究不同浓度的瑞舒伐他汀钙对兔血管平滑肌细胞(Vascular Smooth Muscle Cell,VSMC)转化生长因子β1(Transforming Growth Factorβ1,TGF-β1)表达的影响。方法 培养的兔VSMC分别给予瑞舒伐他汀钙的不同浓度刺激,采用Western Blot法检测细胞中TGF-β1蛋白质的表达量,RT-PCR检测其基因表达量,并且进行半定量比较。结果 免疫印迹显示瑞舒伐他汀钙在10-7~10-3 mol/L的浓度刺激时均显示抑制TGF-β1蛋白表达,刺激12h后,10-7~10-5 mol/L的各组之间随瑞舒伐他汀钙浓度增加TGF-β1蛋白表达量逐渐减少,差异有统计学意义(P〈0.05),继续增加其浓度TGF-β1蛋白表达量不但不再减少,反而有增加趋势。RT-PCR结果显示在刺激3h后,10-5~10-3 mol/L浓度的瑞舒伐他汀钙组TGF-β1mRNA表达量明显低于对照组,差异有统计学意义(P〈0.05),刺激6h以后,10-7~10-3 mol/L浓度的瑞舒伐他汀钙的五个组的TGF-β1mRNA表达量全部低于对照组,差异有统计学意义(P〈0.05)。结论 瑞舒伐他汀钙抑制兔VSMC中TGF-β1中蛋白和基因表达,其表达量可在一定范围内随其浓度的增加而减少。Objective To research the effect of Rosuvastatin on the expression of TGF- β1 in VSMC. Methods The cultured VSMC was simulated by Rosuvastatin with different concentrations. The protein of TGF - β1 was examined by Western Blot and the gene expression was examined by RTPCR. Results The pro- tein expression of TGF- β1 in VSMC was reduced by Rosuvastatin at the concentration of 10^-7 -10^-3mol/ L. After stimulating 12hours,The protein of TGF-β1 in VSMC was reduced with concentration dependent at 10^-7 - 10^-5mol/L. The gene expression of TGF-β1 in VSMC was reduced by Rosuvastatin both at the concentration of 10^-5- 10^-3 mol/L after stimulating 3 hours and at the concentration of 10^-7- 10^-3 mol/L after stimulating 6 hours,there were statistically significant differences (P〈0.05). ConcLusion Rosuvastatin can reduce the protein and gene expresstion of TGF- β1 in rabbit VSMC. And they are concentration dependent within limits.
