摘 要:目的研究不同低温条件下心肌细胞微管的变化。方法常规方法分离SD成年大鼠心肌细胞,-6随机分成4个对照组(单纯KB液保存)和4个实验组(KB液中含紫杉酚,浓度为10 M),利用免疫荧光的方法检测各组在4℃、10℃、15℃、20℃条件下,分别保存4h后的微管网络结构的变化,同时利用生物化学比色法检测保存液中乳酸脱氢酶(LDH)含量。用免疫印迹法检测聚合态微管蛋白的相对含量。结果 4℃、10℃、15℃和20℃温度保存后,微管数量和微管平均光密度值明显高于对照组,LDH释放量比对照组明显降低,而心肌组织内的聚合态微管蛋白含量,随着保存温度的升高逐渐减少。结论心肌组织或单细胞保存的最适温度是4℃。Objective To investigate the changes of cadiomyocytes microtubule structure in different hypothermy. Methods The cadiomyocytes from SD rat were divided randomly into control group(preserved in KB solution) and experimental groups(appending taxol at 10-6 M in KB solution) and preserved for 4h in 4℃ or 10℃ or 15℃ or 20℃ separately. Then microtubular network structure was detected with immunofluorescence, the content of LDH in preservation solution was measured with colorimetry, the expression of polymg α-tubulin was detected by Western blotting. Results The number of microtubules and mean optic density values in cardiomyocytes were increased, but the percentage of released LDH in preservation solution was decreased, in 4℃ or 10℃ or 15℃ preservation groups than in normal control. The relative expression level of polymg α-tubulin in cardiomyocytes was diminished gradually as the preservation temperature increased. Conclusion The optimal temperature is 4℃for the preservation of the cultured cadiomyocytes.
