摘  要:目的探讨缺氧诱导因子1α(HIF-1α)对宫颈癌细胞的生物学行为的影响。方法选用Hela细胞、空载体EGFP-C1转染Hela、EGFP-HIF-1α反义Hela细胞,在37℃,5%CO2,1%O2培养环境中培养细胞对照;通过Western blotting检测各组细胞内HIF-1α蛋白表达,用流式细胞仪检测各组细胞的凋亡率、软琼脂克隆形成方法观察各组细胞的增殖、Transwell侵袭小室方法观察各组细胞的侵袭能力的改变。结果蛋白免疫印迹结果显示,在低氧环境中Hela细胞和仅转染EGFP-C1质粒的Hela细胞中有HIF-1α蛋白的表达条带,而EGFP-HIF-1α反义Hela细胞无表达。并且在低氧环境中EGFP-HIF-1α反义Hela细胞与Hela细胞和仅转染EGFP-C1质粒的Hela细胞相比,其细胞的凋亡率增加、细胞的侵袭能力及细胞集落的形成被抑制,差异均有统计学意义(P<0.01)。结论 HIF-1α能够对宫颈癌Hela细胞的恶性生物学行为产生影响,包括抗凋亡、促增殖、侵袭转移等,且在体外抑制HIF-1α的表达对宫颈癌细胞的凋亡有抑制作用。Objective To explore the direct influence of hypoxia-inducible factor-1α (HIF-1α) on the development of cervical cancer. Methods Cells .were divided into three groups, non-transfected, pEGFP- HIF-1α transfected ,antisense pEGFP- HIF-1α transfected, cultured on the hypoxia (37℃, 5%CO2, 1%O2)conditions, respectively. The expression of HIF-1α was detected by Western blot,flow cytometry and soft agaragar assay, and the transwell cabin assay was employed to evaluated the apoptosis, proliferation and invasion. Results In the hypoxia environment, significantly decreased HIF-1α expression was noted in the anti-HIF-1α-pEGFP-transfected (blocked)Hela cells in hypoxic conditions compared to the blank pEGFP vector-transfected cells, and overall apoptosis was promoted, proliferation and transmembrance behavior inhibited, and the variations was significant (P 〈 0.01 ). Conclusions HIF- 1α may participate in malignant biology behaviors of cervical cancer such as anti-apoptosis, accelerating proliferation and invasion.Suppression of HIF-1α expression in vitro can inhibit cervical cancer cell line.
