摘  要:目的探讨4种主要的细胞色素P450酶在非酒精性脂肪肝(NAFLD)发病进程中的表达变化。方法建立大鼠NAFLD的病理模型,并分别测定肝匀浆中谷草转氨酶(AST)、谷丙转氨酶(ALT)和三酰甘油(TG)等生化指标。采用Western blot法检测在各个病理时期CYP1A2、CYP2D6、CYP2E1和CYP3A1的表达水平的变化。结果随着NAFLD严重程度的增加,AST与ALT的活性显著上升(P<0.01),TG的水平则在重度脂肪肝时期之前上升,而后显著下降(P<0.01)。CYP1A2和CYP2D6的表达水平在发病进程中总体呈下降趋势,CYP3A1虽总体下调,但幅度不大,而CYP2E1的表达水平则明显上升(P<0.01)。结论在NAFLD的发病进程中CYP1A2、CYP2D6、CYP3A1和CYP2E1的表达水平发生明显的改变,实验结果可以为内科医师针对NAFLD患者的用药提供指导。Objective To study the alterations of cytochrome P450 enzymes in rats liver with progressive stages of non-alcoholic fatty liver disease ( NAFLD) . Methods NAFLD models were established. Activities of hepatic as-partate aminotransferase ( AST) , alanine aminotransferase ( ALT) and levels of triglyceride ( TG) were measured by assay kit. The CYP1A2, CYP2D6, CYP2E1 and CYP3A1 protein expressions were detected by Western blot a-nalysis. Results Higher activities of ALT and AST were detected(P〈0. 01)as the severity of NAFLD increased. TG levels increased remarkably in the range from normal to serious steatosis, and then decreased dramatically. Pro-tein expressions of CYP1 A2 , CYP2 D6 and CYP3 A1 tended to decrease with NAFLD progression while CYP2 E1 was increased consecutively. Conclusion These results suggest that significant and novel changes occur in hepatic P450 expression during progressive stages of NAFLD. It may provide a valuable framework for physicians when de-termining the pharmacotherapeutic options and dosing regimens to patients with this disease.
