摘  要:目的优选土党参多糖的硫酸化工艺。方法用三氧化硫-吡啶法对土党参多糖进行硫酸化修饰,以硫酸基取代度作为考察指标,对磺化试剂与多糖质量比、反应温度、反应时间3个因素进行优化;以硫酸钡比浊法测定硫酸基的取代度,红外光谱法测定其结构。结果对取代度影响的顺序为:反应时间>反应温度>磺化试剂与质量比;进行硫酸化修饰时取代度最高可达1.66%,红外光谱谱图显示土党参多糖已硫酸酯化成功。最佳硫酸化条件为:三氧化硫-吡啶与多糖配比为4∶1、反应温度为80℃、反应时间4 h。结论三氧化硫-吡啶法对土党参多糖硫酸化修饰是可行的。OBJECTIVE To optimize the sulfated modification conditions of Campanumoea javanica polysaccharide (CJP). METH- ODS CJP was modified by sulfur trioxide pyridine complex. Substitution degree as the index,L9 (34 ) orthogonal test was used to opti- mize the ratio of sulfur trioxide pyridine complex to CJP, reaction temperature and reaction time. The substitution degree was determined by barium sulfate turbidimetry. Then the structure was analyzed by IR spectrum. RESULTS The order of effect on the substitution degree was the reaction time 〉 reaction temperature 〉 the ratio of sulfonating reagent to polysaccharide. The highest - level of substitu- tion degree reached 1.66%. IR spectrum showed that sulfation of CJP was succeed. The optimal conditions were the ratio of sulfur triox- ide pyridine complex to CJP of 4 : 1, the reaction temperature of 80 ℃ and the reaction time of 4 h. CONCLUSION CJP is modified successfully by sulfur trioxide pyridine complex method.
