摘 要:目的探讨苦参碱对大鼠脑缺血-再灌注损伤神经细胞的保护作用。方法将60只SD大鼠随机分为6组。术前48h,假手术组和手术模型组大鼠ip生理盐水,预处理组ip苦参碱溶液,依达拉奉组注射依达拉奉注射液。除假手术组外,各组建立大鼠全脑缺血-再灌注损伤模型,观察并进行各组大鼠造模后48 h内的神经行为学评分;造模48 h后,计算各组大鼠的脑梗死面积,并取大鼠脑组织,采用比色法检测各组大鼠脑组织中超氧化物歧化酶活性(SOD)及丙二醛(MDA);HPLC法测定谷氨酸(Glu)、门冬氨酸(Asp)的含量,火焰原子吸收法测定Ca2+的含量;采用烘干法测定脑组织的含水量。结果假手术组神经行为学评分无明显变,模型组术后48 h内的神经行为学评分高于假手术组。与模型组比较,苦参碱各预处理组和依达拉奉注射液组神经行为评分均显著降低。与假手术组对比,模型组脑梗死面积明显增大;与模型组相比,苦参碱8、16 mg·kg-1剂量组、依达拉奉组脑梗塞面积均明显下降。与假手术组相比,模型组大鼠脑组织的含水量和Glu、Asp、MDA、Ca2+的含量明显升高,SOD活性明显下降,与模型组相比,各预处理药物能明显降低大鼠脑组织含水量和Glu、Asp、MDA、Ca2+含量,明显升高SOD活性。结论苦参碱对大鼠脑缺血-再灌注损伤具有神经细胞保护作用。Abstract: OBJECTIVE To investigate the protective effect of matrine on ischemia - reperfusion injury of nerve cells. METHODS 60 SD rats were randomly divided into 6 groups. Preoperative 48 h, sham operation group and model group rats were given celiac injection of saline, matrine pretreatment group rats were given matrine injection, Edaravone group rats were given Edaravone. Groups except sham operation group were established model of whole brain ischemia - reperfusion injury in rats, within 48 h after making model, neurobehavioral scores were observed, after 48 h, infarction area in rat was calculated. The super oxide dismutase (SOD) activity and malondialdehyde(MDA) were detected by chemical colorimetry. And glutamate(Glu), Aspartic acid(Asp) were detected by High performance liquid chromatography, content of Ca2+ was detected by flame atomic absorption spectrometry; the content of water in brain was detected by drying method. RESULTS Neural function defect scale of sham operation group had no obvious change, and the postoperative neurobehavioral scores within 48 h of model group were higher than that of control group. Compared with the model group, neurobehavioral scores of matrine pretreatment group and edaravone group were lower with statistically significant. Compared with sham operation group, operation cerebral infarction area of model group significantly increased. Compared with the model group, cerebral infarction area of matrine 8 mg·kg-X dose groups, 16 mg·kg-1 dose group and Edaravone group were significantly lower. Compared with sham operation group, the content of water, Glu, Asp, MDA, Ca2+ of model group increased significantly, while SOD activity decreased obviously. Compared with the model group, the content of water, Glu, Asp, MDA, Ca2 + of matrine pretreatment group obviously reduced, which SOD activity increased significantly. CONCLUSION Matrine show cerebral protection in ischemia - reperfusion injury.
