摘 要:目的:探讨树舌多糖GF对人肝癌细胞MHCC97H荷瘤裸鼠肿瘤生长的影响及机制。方法:人肝癌细胞株MHCC97H接种4~5周鼠龄的雄性BALB/c-nu裸鼠背部2周,建立裸鼠皮下瘤模型。30只荷瘤裸鼠随机分为3组:对照组、生理盐水组及树舌多糖GF组,每组10只。对照组未做处理;生理盐水组腹腔注射生理盐水(0.15ml/只),连续14天;树舌多糖GF组按50mg/kg,0.15ml/只腹腔注射树舌多糖GF,连续14天。于末次给药24h,处死全部裸鼠,解剖瘤体,比较各组肿瘤体积及重量。Western blot检测各组瘤体Notch细胞信号的胞内段(Notch intracecellar domain,NICD)蛋白的表达情况。结果:给药14天后,树舌多糖GF组的瘤体及瘤重分别为(81.21±25.68)mm3和(1.39±0.41)g,与对照组(1022.14±164.71)mm3和(2.30±0.46)g,以及生理盐水组(983.19±114.27)mm3和(2.11±0.58)g相比较均有统计学差异(P〈0.05),树舌多糖GF可抑制肿瘤生长及减少瘤重;Western blot结果显示相比对照组及生理盐水组,树舌多糖GF组NICD表达降低。结论:树舌多糖GF抑制人肝癌细胞MHCC97H荷瘤裸鼠肿瘤生长,其抗肿瘤的作用靶点之一可能为Notch细胞信号。Objective:To investigate the effect of ganoderma appanatum polysaccharider GF on the xenograft tumor growth of nude mice models with human MHCC97H hepatocellular carcinoma cell line. Methods:Human MHCC97H hepatocellular carcinoma cell was injected into subcutaneous layers of the back of nude mice for es- tablishing subcutaneous xenograft tumor models. After 2 weeks, thirty subcutaneous xenograft tumor model mice were randomly divided into three groups:a control group (no treatment), a mock group (abdomen injec- ted normal saline) and a treatment group (abdomen injected ganoderma appanatum polysaccharider GF). Af- ter the treatment for 14 days, all the mice were killed and compared according to tumor size and weight, and Notch intracecellar domain (NICD) protein expressions in three groups were detected by Western blot. Re- sults:After the treatment for 14 days, the tumor volume and weight in the treatment group were (81.21 ± 25.68 )mm3and( 1.39 ±0.41 )g, which was significantly different compared with the control group (1022.14 ± 164.71)mm3and (2.30 ±0.46)g, and the mock group(983.19 ±114.27) mm3and (2.11 ±0.58)g in statistical analysis (P 〈 0.05). Tumor was inhibited by ganoderma appanatum polysaccharider GF. Compared with the control group and the mock group, the NICD protein expression of treatment group was decreased. Conclusion:Ganoderma appanatum polysaccharider GF can inhibit the xenograft tumor growth of nude mice models, and its antitumor mechanism is related to Notch cell signal.
