电针傍刺对大鼠皮肤压疮Ang-1及Ang-2表达的影响

摘　　要：目的:研究电针傍刺对大鼠皮肤压疮血管生成素Ang-1及Ang-2表达的影响,探讨电针傍刺对大鼠皮肤压疮修复的作用及作用机制。方法:90只SD大鼠采用随机数字表法分成电针组、针刺组与空白组,每组30只。再将其随机分入5个治疗时间点,即1天组、3天组、5天组、7天组、9天组,每小组6只。在大鼠腿部近膝关节骨隆突处用造模装置制造压疮后针刺组取压疮局部皮肤进行傍刺治疗,电针组在针刺组基础上配合电针治疗,每次30min,每日1次。连续观察治疗过程中创面/修复的形态学变化并应用免疫组化法检测Ang-1与Ang-2的表达情况。结果:大鼠压疮皮肤Ang-1表达阳性细胞累计积分,电针3天组与9天组均高于针刺与空白3天与9天组(P<0.05);电针3天组高于电针1天组(P<0.05);针刺5天组低于针刺3天组(P<0.05);大鼠压疮皮肤组织Ang-2表达阳性细胞累计积分,电针1天组与9天组均高于空白1天组与9天组(P<0.05);电针3天组与7天组均高于针刺与空白3天与7天组(P<0.05);电针5天组高于针刺5天组(P<0.05);针刺5天组高于空白5天组(P<0.05);电针3天组高于电针1天组(P<0.05);电针5天组低于电针3天组(P<0.05);针刺7天组低于针刺5天组。结论:电针傍刺能促进大鼠压疮皮肤创伤修复,其机制可能与调节不同时间内压疮皮肤中Ang-1及Ang-2表达情况有关。Objective ：To study the effect of Bangci electroacupuncture on the expression of Angiogenin - 1 and Angiogenin- 2 in pressure ulcer rats and explore the effect and mechanism of Bangci electroacupuneture on the restoration of pressure sores. Methods ： Ninety SD rats were divided into an EA group, an acupuncture group and a blank group with randomized digital table. Each group had 30 rats. Then they were randomly divided in- to 5 treatment time, ld group,3d group, 5d group, 7d group and 9d group,with 6 rats in each case. Make pressure sores by applying the modeling apparatus on the rats＇ legs, which were near the bone protuberance of the knees. Then the acupuncture group was treated by Bangei to treat the local pressure sores and the EA group was treated by Bangci EA, 30 rain per time, once a day. Continuously observe the morphologic varia-tions in the process of sores restoring and apply immunohistochemical method to measure the expression of Ang - 1 and Ang -2. Results：The positive cell accumulated score of Ang - 1 expression in the EA 3d and 9d groups was higher than that in the blank 3d and 9d groups in pressure ulcer rats （ P 〈 0.05 ）. The positive cell accumulated score of Ang - 1 expression in the EA 3d group was higher than that in the EA ld group in pres- sure ulcer rats （P 〈 0.05 ）. The positive cell accumulated score of Ang - 1 expression in the acupuncture 5d group was lower than that in the acupuncture 3d group in pressure ulcer rats （P 〈 0.05 ）. The positive cell ac- cumulated score of Ang -2 expression in the EA ld and 9d groups was higher than that in the blank ld and 9d groups in pressure ulcer rats （ P 〈 0.05 ）. The positive cell accumulated score of Ang - 2 expression in the EA 3d and 7d groups was higher than that in the acupuncture 3d and 7d groups in pressure ulcer rats （P 〈 0. 05 ）.
