摘 要:目的:探讨小檗碱对腹腔暴露后大鼠肠黏膜屏障的影响。方法:将大鼠剖腹后暴露腹腔,但不进行任何手术操作,暴露3 h后关腹,制作腹腔暴露模型。将30只大鼠随机分为五组,即对照组、腹腔暴露组、小檗碱低剂量(100 mg/kg)干预组、中剂量(150 mg/kg)干预组和高剂量(200 mg/kg)干预组,每组6只。小檗碱低、中、高剂量干预组分别于术前3 d和术后6 h每天给予一次相应剂量的小檗碱灌胃。对照组和腹腔暴露组在相同条件下分别给予等量的等渗盐水灌胃。术后24 h取血检测血清D-乳酸水平,并取末端回肠组织测定肠道FD4清除率,同时观察末端回肠组织的病理变化,并进行肠黏膜损伤Chiu's评分。结果:与对照组比,腹腔暴露组大鼠血清D-乳酸水平明显上升(P<0.05),肠道FD4清除率和肠黏膜损伤Chiu's评分明显增加(P<0.05)。与腹腔暴露组比,各小檗碱干预组大鼠血清D-乳酸水平有所下降,肠道FD4清除率和肠黏膜损伤Chiu's评分有所降低,以中、高剂量组最为显著(P<0.05)。结论:小檗碱可减轻腹腔暴露引起的肠黏膜损伤,对维持肠黏膜屏障功能具有一定的保护作用。Objective : To investigate the effect of berberine on intestinal mucosa barrier in rat in- duced by peritoneal air exposure. Methods : The laparotomy was used, without any surgical procedure, but with peritoneal air exposure for 3 h, to induce the model for peritoneal air exposure. Thirty rats were randomized into five groups:a control group, a peritoneal air exposure model group, and three berberine treatment groups with low (100 mg/kg) , moderate (150 mg/kg) and high dose (200 mg/kg), respec- tively ( n = 6 each). At 24 h after surgery, blood was obtained to determinate serum D-lactate, terminal ileum was sampled to determinate intestinal clearance of FD4 and to observe histological changes and Chiug score for damage of intestinal mucosa. Results: Compared with the control group, the model group showed a significant increase in serum D-lactate, intestinal clearance of FD4 and Chiu's score for damage of intestinal mucosa ( P 〈 0. 05 ). Compared with the model group, three berberine treatment groups showed different degrese of decrease in serum D-lactate, intestinal clearance of FD4 and Chiu's score. exposure Conclusion: The berberine could decrease intestinal mucosal damage induced by peritoneal air and further maintain intestinal mucosal barrier.
