摘 要:目的探讨不同强度运动对大鼠心肌线粒体解偶联蛋白2(UCP2)表达的影响及其与心肌能量变化的关系。方法雄性Wistar大鼠45只,随机分为安静对照组,渐进训练组、渐进训练力竭组,每组15只。4周后,测定血清游离脂肪酸(FFA)浓度及心肌线粒体三磷酸腺苷(ATP)含量,反转录聚合酶链反应(RT-PCR)及Western blotting测定心肌线粒体UCP2的表达变化。结果渐进训练力竭组ATP含量明显低于安静对照组与渐进训练组(P<0.01);渐进训练力竭组血清FFA浓度明显升高,分别是安静对照组和渐进训练组的2.0倍和1.7倍(P<0.01)。RT-PCR及Western blotting结果显示,与安静对照组及渐进训练组相比,渐进训练力竭组心肌线粒体UCP2 m RNA和蛋白表达明显升高(P<0.01)。相关性分析显示,心肌线粒体UCP2的表达与血清FFA浓度呈正相关(r=0.795,P<0.01),而与ATP含量呈负相关(r=-0.843,P<0.01)。结论力竭运动训练可引起大鼠血清FFA浓度升高及心肌线粒体ATP含量降低,同时可诱导心肌线粒体UCP2表达。UCP2的表达与FFA浓度呈正相关,而与ATP含量呈负相关,提示UCP2可能参与了力竭运动中心的肌能量代谢。Objective To study the effect of different exercise intensity on the expression of uncoupling protein 2 (UCP2) in mitochondria of rat myocardium, and investigate the correlation and significance between energy metabolism and UCP2. Methods Forty five male Wistar rats were randomly divided into 3 groups (15 each): normal control (NC) group, endurance exercise (EnE) group and exhaustive exercise (ExE) group. Four weeks later, both the serum concentration of free fatty acid (FFA) and the content of ATP in myocardial mitochondria were measured. The expression of UCP2 in myocardial mitochondria was assessed with RT-PCR and Western blotting. Results The content of ATP was lower in ExE group than that in NC and EnE group (P〈0.01). The serum FFA concentration in Ex.E group was 2.0 folds and 1.7 folds higher than in that of NC group and EnE group, respectively (P〈0.01). The results of RT-PCR and Western blotting indicated that the expression of UCP2 in myocardial mitochondria was significantly up-regulated in ExE group as compared with that of NC and EnE groups (P〈0.01). Correlation analysis showed a positive correlation between expression of UCP2 and serum FFA concentration (r=0.795, P〈0.01), while a negative correlation was found between expression of UCP2 and ATP content (r=-0.843, P〈0.01). Conclusions Exhaustive exercise may increase serum FFA concentration and decrease myocardial mitochondria ATP content, and it may induce myocardial mitochondria UCP2 expression at the same time. UCP2 expression is positively correlated with FFA concentration, but it was negatively correlated with the ATP content, implying that UCP2 may participate in the process of myocardial energy metabolism during exhaustion exercise.
