摘 要:目的探讨趋化素样因子1(CKLF1)及其拮抗肽C19对不同时期特应性皮炎(AD)患者外周血T细胞亚群的作用。方法应用淋巴细胞分离液分离急性期和慢性期AD的外周血单个核细胞(PBMC),运用流式细胞术(FACS)检测CKLF1在其外周血T细胞亚群中的表达,并与正常对照组比较;体外诱导培养AD患者的Th1,Th2细胞亚群,以不同浓度CKLF1和(或)C19刺激上述细胞,并采用酶联免疫吸附法(ELISA)检测培养上清中IFN-γ,IL-4,IL-17水平变化。结果急性期AD患者Th2细胞中CKLF1的表达高于对照组(P<0.05),慢性期AD患者Th1细胞中CKLF1的表达高于对照组(P<0.05)。AD患者外周血CKLF1在Th17细胞中表达与正常对照组间差异无统计学意义。急性期AD患者体外培养的Th2细胞产生的IL-4水平高于对照组(P<0.05),慢性期AD患者体外培养的Th1细胞产生的IFN-γ水平高于对照组(P<0.05)。IL-17在急性期和慢性期AD患者体外培养的Th1细胞中的水平与正常对照组相比差异均无统计学意义。急性期AD患者的Th2细胞经CKLF1刺激,产生IL-4水平高于未刺激组(P<0.05)。经CKLF1和C19(10ng/m L)共同培养后,IL-4的水平降低。慢性期AD患者的Th1细胞经CKLF1刺激,产生IFN-γ的水平高于未刺激组(P<0.05)。经CKLF1和C19(10ng/m L)共同培养后,IFN-γ的水平无显著改变。结论 CKLF1在AD患者发病中可能起着重要的病理性作用,CKLF1的拮抗肽C19可能对急性期AD患者具有潜在治疗作用。Objective To explore the influence of chemokine - like factor 1 ( CKLF1 ) and CKLF1 antagonism peptides C19 on AD related T cells. Methods Peripheral blood mononuclear cells (PBMC) were separated from acute atopic dermatitis patients, chronic atopic dermatitis patients and healthy controls. The expression levels of CKLF1 in peripheral blood T cell subsets were detected by FACS. ELISA were used to detect IFN-γ, IL- 4 and IL-17 levels secreted by the peripheral Thl cells or Th2 cells which had been stimulated by different concentration of CKLF1 and/or C19, respectively. Results The expression of CKLF1 in Th2 cells from the patients in acute AD was significantly higher than that in control. In the chronic AD, the expression of CK- LF1 in Thl cells was significantly higher than that in control ( P 〈 0. 05 ). The CKLF1 expression in Thl7 cells of AD patients was similar to that of healthy controls ( P 〉 0. 05 ). IL - 4 level in cultured Th2 cells from acute AD patients was significantly higher than that from controls ( P 〈 0.05 ). IFN - γ level in cul- tured Thl cells from chronic AD patients was significantly higher than that from controls ( P 〈 0. 05 ). No statistical difference in IL-17 levels in cultured Thl cells between AD patients and normal control (P 〉 0. 05). IL-4 levels in Th2 cells stimulated with CKLF1 were markedly higher in acute AD patients than in controls (P 〈 0. 05 ). IL- 4 level was significantly decreased when the Th2 cells were cultured with both CKLF1 and C19 (10ng/mL). IFN - γ level in chronic AD was significantly higher than that in controls when Thl cells were stimulated by CKLF1 ( P 〈 0.05 ). IFN -γ level was not markedly decreased when the Thl cells were cultured with both CKLF1 and C19 (10ng/mL). Conclusion CKLF1 may play an important pathological role in AD development. The CKLF1 antagonism peptides C19 may have potential therapeutic effect on patients with acute AD.
