摘 要:目的:利用H-E染色法和Masson染色法对慢性阻塞性肺病(Chronic Obstructive Pulmonary Disease,COPD)大鼠模型进行染色观察。方法:SPF级雄性SD大鼠20只,随机分为实验组和对照组,各10只。实验组大鼠置入自制熏烟染毒箱,熏过滤嘴香烟烟雾(烤烟型)。对照组大鼠不做处理。3个月后处死大鼠,取肺部组织。通过石蜡切片技术、H-E染色、Masson染色对COPD大鼠模型切片进行染色观察。结果:H-E染色显示炎细胞浸润支气管平滑肌层、肺泡壁增厚等现象。Masson染色显示肺泡壁早期纤维化形成等现象。结论:H-E染色法和Masson染色法相结合可较全面的对COPD大鼠模型的病理现象进行观察分析。Objective To observe the rat model of Chronic Obstructive Pulmonary Disease(COPD) by using H-E staining and Chronic staining. Methods20 SPF male SD rats,were randomly divided into experimental group and control group, 10 rats in each group. The rats of the experimental group in smoke exposure box which made by ourselves, smoked cigarette smoke(40mins, a flue-cured tobacco type). Rats in control group was not treated. After 3 months, rats were killed and took lung tissues. The rat model of COPD staining was observed by paraffin section, H-E staining and Masson staining. Results H-E staining showed that inflammatory cell infiltration in bronchial smooth muscle layer, alveolar wall thickening and so on. Masson staining showed that the early stage of pulmonary fibrosis was formed. Conclusion H-E staining method and Masson staining method can be more comprehensive to observe and analyze the pathological phenomena of COPD rat model.
