摘 要:为了研究柑橘黄龙病菌是否存在遗传分化现象,对采自湖南、江西、福建等3个省的20多个县(市)的柑橘黄龙病样品,提取了总DNA,并对16S r DNA和M94319基因序列进行了PCR扩增、测序,通过MEGA5.0生物信息学软件对这些序列进行了分析。结果表明,在柑橘溃疡病遗传多样性分析过程中M94319基因序列的分辨率比16S r RNA高,即M94319基因序列基因片段能有效地检测出黄龙亚洲种,16S r RNA区分不了各个小种。In order to study whether citrus Huanglongbing pathogen has the phenomenon of genetic differentiation, samples with citrus Huanglongbing were collected from more than 20 counties (cities) in Hunan, Jiangxi and Fujian, the total DNA was extracted, 16S rDNA and M94319 gene sequence were amplified by PCR and sequenced, and the sequencing results were analyzed with the bioinformatics software MEGA5.0. The results showed that M94319 gene sequence had higher resolution than 16S rDNA in the analysis of genetic diversity, which indicated that the fragment ofM94319 gene sequence could effectively detect Huanglong Asian species, and 16S rDNA could not distinguish races.
