摘 要:探讨了用BSA-Au NCs荧光体系,并用该体系建立测定氯霉素(chloramphenicol,CAP)的新方法。实验表明,BSA-Au NCs荧光体系在pH=7.6的PBS缓冲溶液中可以发出较强的荧光,而加入CAP后,BSA-Au NCs体系的荧光发生猝灭。且CAP浓度在4.0×10^-9~4.4×10^-8 mol/L范围内与BSA-Au NCs体系荧光猝灭的程度呈良好的线性关系,相关系数r=0.9972,检出限为1.2×10^-9 mol/L(n=11),以此建立利用荧光猝灭法测定CAP的新方法。实验考察了常见离子等对该体系的影响,结果表明这些离子对实验的干扰有限,不会对实验产生影响。新方法应用于牛奶中CAP的检测,其相对标准偏差(RSD)≤1.88(n=6),回收率为98.8%~101.75%。A new fluorescence method for the determination of chloramphenieol(CAP) was developed based on BSA-Au NCs fluorescence probe. It was found that under the conditions of PBS buffer medium at pH 7.6, BSA-Au NCs can emitted higher fluorescence, upon the addition of CAP, the fluorescence quenching of BSA-Au NCs occurred. The change of the BSA-Au NCs fluorescence intensity was in good linear rela- tionship with the concentration of CAP. Under optimum condition, the determination range of CAP was from 4.0×10^- 9~ 4.4×10^-8 mol/L, with a detection limit of 1.2 ×10^- 9 mol/L (n= 11). A novel fluores- cence quenching platform for determination of CAP was thus constructed. Besides, most of common rele- vant substance, cations and anions did not interfere with the detection of CAP. The novel platform has been applied in the determination of CAP in milk examples, results show that the recovery in this BSA-Au NCs method is sufficient, in the range of 98.8~101.75%, with the relative standard deviation(RSD) less than 1.88 %. Above results suggest that the proposed method has a relatively high sensitivity and is good enough in the measuring of CAP.
