摘  要:以壳聚糖为絮凝剂对川牛膝的总多糖进行了絮凝纯化研究。通过单因素考察及正交试验得到优化的工艺条件为:絮凝温度45℃、壳聚糖用量0.4 m L/g生药量、絮凝时间2 h、药液浓缩比1:12、药液p H=4。此时,多糖保留率76.32%、脱蛋白率64.25%,与传统醇沉工艺相比,絮凝法得到的多糖保留率和纯度较高。The essay was aimed at studying flocculating purification of total polysaccharides by using chitosan. The optimum conditions were obtained through single factor and orthogonal experiment. The results were as follows:the temperature was 45 ℃ , dosage of chitosan was 0.4 mL/g raw material, flocculation time was 2 h, solution concentration ratio was 1:12, pH of solution was 4. Under this condition, polysaccharides retention rate was 76.32%, deproteinizing rate of 64.25%. Compared with the traditional alcohol precipitation process, the retention rate and purity of the polysaccharide of the flocculation purification process are higher.
