Effect of Zuogui Wan(左归丸) on Fetal Rats Birth Weight of Gestational Diabetes Mellitus Rats

摘  要:目的观察左归丸对妊娠糖尿病大鼠胎鼠出生体重的影响及可能作用机制。方法 SD大鼠受孕后随机分为正常对照组、妊娠糖尿病组、胰岛素干预组、左归丸干预组、联合干预组,每组10只。受孕当日除正常对照组外其余各组大鼠腹腔注射链尿佐菌素45 mg/kg建立妊娠糖尿病大鼠模型。于妊娠第3天开始左归丸干预组、联合干预组给予左归丸10 g/(kg·d)灌胃,正常对照组和妊娠糖尿病组灌胃等量蒸馏水,每日1次;胰岛素干预组、联合干预组给予诺和灵R 20 IU/(kg·d)皮下注射,每日1次,至孕18天。孕19天解剖大鼠,称胎鼠体重,检测血清、皮质酮、胎盘11β-羟基类固醇脱氢酶2(11β-HSD2)mRNA和蛋白表达。结果与正常对照组比较,妊娠糖尿病组大鼠血清皮质酮水平明显升高,胎鼠体重、胎盘11β-HSD2 mRNA和蛋白表达降低(P<0.05或P<0.01);与妊娠糖尿病组比较,胰岛素干预组、联合干预组大鼠血清皮质酮水平均明显降低,胎鼠体重升高(P<0.05或P<0.01),左归丸干预组的胎鼠体重比较差异无统计学意义(P>0.05),联合干预组胎盘11β-HSD2 mRNA和蛋白表达明显升高(P<0.05)。结论左归丸可以改善妊娠糖尿病病理状态对胎鼠体重的影响,联合胰岛素效更佳,其机制可能是通过上调11β-HSD2的表达、降低皮质酮浓度有关。Objective To observe the effect and gestational diabetes mellitus (GDM) rats. Methods possible mechanism of Zuogui Wan on fetal rats birth weight of Pregnant SD rats were randomly divided into normal control group, GDM group, insulin intervention group, Zttogui Wan intervention group and combined intervention group, 10 rats in each group. On the day of conception, rats of these groups were injected intraperitoneally with streptozocin 45 mg/kg to establish GDM rat models except rats of normal control group. From the third day of conception, Zuogui Wan intervention group and combined intervention group were given Zuogui Wan 10g/ (kg · d) by gavage. Normal control group and GDM group were given equivalent distilled water by gavage, once each day. Insulin intervention group and combined intervention group were given Novolin R 20IU/ (kg · d) by subcutaneous injection, once each day, until 18 days of conception. On the 19th day of conception, rats were anatomized. Weigh fetal rats, detect serum corticosterone, placenta 11 β-hydroxysteroid dehydrogenase 2 (11β-HSD2 ) mRNA and the expression of protein. Results Compared with normal control group, serum corticosterone in GDM group increased; fetal rats weight, pla- eental1β-HSD2 mRNA and the expression of protein decreased ( P 〈 0. 05 or P 〈 0.01 ). Compared with GDM group, serum corticosterone in insulin intervention group and combined intervention group decreased; fetal rats weight increased ( P 〈 0. 05 or P 〈 0. 01 ). Fetal rats weight in Zuogui Wan intervention group had no significant difference ( P 〉 0. 05 ). Placental 1β-HSD2 mRNA and the expression of protein in combined intervention group increased ( P 〈 0. 05). Conclusion Zuogui Wan could improve the effect of GDM pathological state on fetal rats weight. It was more effective to combine with insulin. The mechanism may relate to up-regulate the expression of 1113-HSD2 and decrease the concentration of corticosterone.
