摘  要:[目的]本文旨在揭示RFRP-3基因在不同年龄段湖羊雄性生殖器官发育中的表达变化规律及其参与繁殖营养调控的作用。[方法]利用荧光定量PCR法检测RFRP-3在10日龄、4月龄、8月龄等不同阶段雄性湖羊(n=3)的睾丸、附睾头、附睾体、附睾尾及输精管中的表达情况;利用免疫组化法检测RFRP-3在8月龄雄性湖羊生殖器官的定位表达;利用荧光定量PCR法分析不同营养饲喂水平下RFRP-3基因在湖羊雄性生殖器官中的表达差异。[结果]RFRP-3基因在睾丸和输精管中相对高表达,在附睾头,附睾体,附睾尾中相对低表达;RFRP-3蛋白分布于睾丸的生精小管内部、输精管及附睾,可见免疫阳性反应;在高能高蛋白水平饲喂下,RFRP-3基因在雄性湖羊生殖器官中表达量显著低于正常饲喂水平。[结论]RFRP-3在湖羊雄性生殖器官内表达,且在不同组织和不同营养饲喂水平下表达量差异显著,为研究RFRP-3的生物学功能及其参与动物繁殖营养调控提供了依据。[Objectives]The aim of the paper is to reveal the difference of RFRP-3 gene expression in the male reproductive organ development of Hu sheep. [Methods]This study examined RFRP-3 expression in testis,epididymis head,cauda epididymis and ductus deferens of Hu sheep,ranging from 10 days,4 months and 8 months( n = 3) respectively through fluorescence quantitative PCR. Immunohitochemical method was used to study the positioning expression of RFRP-3 in reproductive organ of 8 months male Hu sheep. In addition,we analysed the difference of RFRP-3 expression in the male reproductive organs of different feeding level sheep by fluorescence quantitative PCR. [Results]RFRP-3 gene was highly expressed in the testis and vas deferens,but lowly expressed in the epididymis head,epididymisand cauda; RFRP-3 protein was expressed in testicular seminiferous tubule internal,ductus deferens and epididymis. RT-PCR results showed that RFRP-3 expression in the high-energy and high-protein feeding sheep,was significantly lower than that of normal feeding sheep. [Conclusions]All the results showed that RFRP-3 was expressed in the male reproductive organs of sheep,and the expression of RFRP-3 in different groups and different nutrition levels was significant and also provided basis for the research of RFRP-3 and its biological function in nutrition regulation of animal breeding.
