摘  要:目的研究Rho/Rho激酶的底物-肌球蛋白磷酸酶靶亚1(MYPT-1)及缝隙连接蛋白40(Cx40)在房颤患者左心耳中的表达,探讨Rho/Rho激酶信号通路在房颤的发生和维持中的作用。方法 40例接受开胸手术者分为房颤组和窦性心律组,手术中取约100 mg左心耳组织。采用免疫组化ABC法、Western Blot技术检测MYPT-1、Cx40在左心耳组织的蛋白表达水平,并分析MYPT-1、Cx40在房颤组中的相关性。结果免疫组化和Western Blot结果均显示,房颤组患者MYPT-1、Cx40在左心耳中的表达明显高于窦性心律组(P<0.01)。相关分析显示,房颤组MYPT-1与Cx40的蛋白表达呈显著正相关(r=0.989,P<0.01)。结论房颤患者左心耳组织中明显存在MYPT-1表达上调与功能活化,提示Rho激酶信号转导通路在房颤病变中被激活,其机制可能因调节Cx40而发挥作用。Objective To explore the expressions of myosin phosphatase target subunit- 1( MYPT- 1) and connexin 40( Cx40) in left atrial appendage of patients with atrial fibrillation( AF). Methods Left atrial appendage tissue samples of 40 patients undergoing cardiac surgery were examined. Among the patients examined,20 had AF,20 had no history of AF. The expressions of MYPT- 1 and Cx40 in left atrial appendage were detected by immunohistochemistry and western blot. Results Immunohistochemistry and western blot showed that the expressions of MYPT- 1 and Cx40 were significantly up- regulated in left atrial appendage of patients with AF than patients with sinus rhythm( P〈0. 01).The two indicators were positively correlated( r = 0. 989,P〈0. 01). Conclusion The MYPT- 1 expression in left atrial appendage of patients with AF is up- regulated and activated,MYPT- 1 promotes the expression of Cx40,which suggests that Rho kinase signal transduction may play an important role in the pathological changes in AF.
