摘  要:[目的]为利用百合未减数花粉进行有性多倍化育种。[方法]利用流式细胞仪测定百合花粉的倍性,以Marlon、Viviana的花粉为材料,以叶片为对照,比较两种不同细胞核提取液的效果差异。[结果]Cy Stain UV Precise T提取液明显优于WPB提取液;Marlon、Vivian叶片测定结果显示其为二倍体,花粉测定的图像显示两个品种都能自发产生2n花粉。[结论]流式细胞仪可以快速地测定百合花粉的倍性。[ Objective ] For the use of lihium unreduced pollen to sexual polyploidization breeding. [ Method ] We made use of flow cytometry to determine lilium pollen ploidy by taking the pollen of Marion and Viviana as materials and leaves for comparison. [ Result ] By comparing the effect difference of two different cell nuclear extracts, the extract of CyStain UV Precise T was significantly better than that of WPB. Marion and Viviana leaves measurement results showed that they were diploids and the pollen measurement image showed the two varieties could produce 2n pollen on their own. [Conclusion] Flow cytometry can be used to quickly measure lilium pollen ploidy.
