摘 要:DQB1基因在抗原呈递和免疫系统中发挥重要的作用。为了研究河西绒山羊DQB1基因的核苷酸序列变异和跨种进化,采用PCR-SSCP和测序技术,检测899只河西绒山羊DQB1基因的多态性,并用来自山羊、绵羊和黄牛DQB1基因的33条核苷酸序列构建NJ树。结果表明,河西绒山羊DQB1基因存在6个等位基因和34个核苷酸变异位点,变异位点数目占分析位点数目的15.53%。经与GenBank中山羊DQB1基因的核苷酸序列比对后发现,5个等位基因为首次发现。系统进化研究表明,山羊DQB1基因的所有核苷酸序列均聚在第1支中,部分核苷酸序列与绵羊、牛的序列相似性高于山羊之间的序列相似性。因此,河西绒山羊DQB1基因多态性丰富,在山羊、绵羊和牛中存在跨种多态性。DQB1 gene plays an important role in antigen-presenting and immune system. To study allelic variation and trans-species evolution in DQB1 gene in Hexi cashmere goat, eight hundred and ninety-nine goats from Hexi cashmere goats were used to detect polymorphism of DQB1 gene by PCR-single-strand conformational Polymorphism (SSCP) followed by DNA sequencing analysis, and 33 nucleotide sequences from goat, sheep and cattle were used to construct NJ tree. A total of six unique alleles of DQB1 and 34 nucleotide polymorphism sites, which occupied 15. 53% of whole exon 2 sequences, were identified in Hexi cashmere goats. By sequence alignment to all DQB1 sequences of goat in GenBank, five nucleotide sequences were newly found in the study. Phylogenetic analysis revealed that all caprine sequences clustered into the first clade, and some of our identified cashmere goat DQB1 sequences were more similar to sheep and cattle than other goat sequences. In conclusion,DQB1 gene is polymorphic in Hexi cashmere goat and trans-species polymorphism was found in goat, sheep and cattle.
