摘  要:目的研究蝎毒活性肽对同型半胱氨酸引起的血管内皮细胞损伤的蛋白表达谱的变化及其蛋白质组学机制。方法用二向电泳、图像分析、基质辅助激光解析电离化/飞行时间质谱鉴定等蛋白质组学技术测定同型半胱氨酸致血管内皮细胞损伤表达量有显著改变的蛋白质,Western blot测定血管内皮细胞锰超氧化物歧化酶、网钙蛋白1前体和钙网蛋白前体蛋白的表达。结果与空白对照组比,同型半胱氨酸组发现有11个蛋白点下调及1个蛋白点上调。与同型半胱氨酸组比,同型半胱氨酸+蝎毒活性肽组上调锰超氧化物歧化酶和网钙蛋白1前体,下调钙网蛋白前体的蛋白表达。结论靶蛋白的发现说明蝎毒活性肽可抵抗由高同型半胱氨酸引起氧化应激造成的血管内皮细胞损伤。AIM To study the effects of scorpion venom active peptides( SVAP) on the changes of protein expression spectrum in vascular endothelial cell injury induced by homocysteine and their mechanisms of proteomics. METHODS The changes of protein expression of vascular endothelial cell injured by homocysteine were determined by bi-directional electrophoresis,image analysis,MALDI-TOF-MS. Superoxide dismutase [Mn ],reticulocalbin 1 precursor and calreticulin precursor in vascular endothelial cells injured by homocysteine were determined with Western blot. RESULTS Compared with the control group,eleven protein spots were down-regulated and one protein spot was up-regulated in homocysteine group. Compared with homocysteine group,homocysteine + SVAP could up-regulate the protein expressions of the superoxide dismutase [Mn]and eticulocalbin 1 precursor and down-regulate the expression of calreticulin precursor. CONCLUSION Target proteins reveal that SVAP has protective effect on vascular endothelial cell injury induced by oxidative stress.
